摘 要:目的 探索畅枢汤在大鼠痤疮模型中,对TLR4/TNFa/IL-1信号通路和角化标志物CK10的影响。方法 构建大鼠痤疮模型,实验组给予畅枢汤7.71 g/(kg·d)灌胃给药,对照组分别给予异维A酸胶囊3.125 mg/(kg·d)及畅枢汤+异维酸胶囊灌胃给药,设立空白组和模型组。连续干预6周,组织标本取材,提取总RNA及总蛋白。免疫组化、qRT-PCR和WB检测TLR4、NF-κB、IL-1、CK10的表达。结果 大鼠痤疮模型构建成功。TLR4、NF-κB、IL-1、CK10基因及蛋白在各组之间有不同程度的表达,模型组的表达量最高,各药物干预组表达量较模型组明显下降,异维A酸联合畅枢汤组表达量下降最明显。畅枢汤组(TLR4:0.20±0.03,NF-κB:0.18±0.04,IL-1:0.16±0.01,CK10:0.15±0.02)明显低于模型组(TLR4:0.25±0.04,NF-κB:0.23±0.02,IL-1:0.21±0.03,CK10:0.18±0.03)(P<0.05),高于空白对照组(TLR4:0.09±0.02,NF-κB:0.11±0.02,IL-1:0.11±0.01,CK10:0.08±0.01)和畅枢汤与异维A酸胶囊联合用药组(TLR4:0.12±0.01,NF-κB:0.13±0.01,IL-1:0.12±0.01,CK10:0.11±0.01)(P<0.05)。结论 大鼠痤疮模型中,畅枢汤可降低TLR4、NF-κB、IL-1的含量、下调CK10的表达;同时与异维A酸胶囊联合时,TLR4、NF-κB、IL-1含量的降低和CK10的表达下调更为显著;初步推测畅枢汤是通过抑制TLR4/TNFa/IL-1信号通路关键成员,进而下调CK10的表达。Objective To explore the effect of Changshu Decoction on TLR4/TNFa/IL-1 signaling pathway and keratinization marker CK10 in rat acne model.Methods The rat acne model was established.The experimental group was given Changshu Decoction 7.71 g/(kg·d) by gavage,and the control group was given isotretinoin capsule 3.125 mg/(kg·d) and Changshu Decoction + isotretinoin capsule by gavage,respectively.The blank group and model group were assigned.After 6 weeks of continuous intervention,tissue samples were taken to extract total RNA and total protein.Expressions of TLR4,NF-κB,IL-1 and CK10 were detected by immunohistochemistry,qRT-PCR and WB.Results The rat acne model was successfully constructed.TLR4,NF-κB,IL-1,CK10 genes and proteins were expressed in different degrees among the groups.The expression level in the model group was the highest.The expression level in each drug intervention group was significantly lower than that in the model group.The expression level in the group of isotretinoin combined with Changshu decoction was the most significant,and it was significantly lower in Changshu Decoction group(TLR4:0.20±0.03,NF-κB:0.18±0.04,IL-1:0.16±0.01,CK10:0.15±0.02) than that in model group(TLR4:0.25 ±0.04,NF-κB:0.23±0.02,IL-1:0.21 ±0.03,CK10:0.18 ±0.03)(P <0.05),and higher than that in blank control group(TLR4:0.09±0.02,NF-κB:0.11±0.02,IL-1:0.11±0.01,CK10:0.08±0.01) and the group of Changshu decoction combined with isotretinoin capsules(TLR4:0.12 ±0.01,NF-κB:0.13 ±0.01,IL-1:0.12 ±0.01,CK10:0.11 ±0.01)(P <0.05).Conclusion In the rat acne model,Changshu Decoction can reduce the contents of TLR4,NF-κB and IL-1,and downregulate the expressions of CK10;at the same time,when combined with isotretinoin capsules,the contents of TLR4,NF-κB and IL-1 decrease.The expressions of CK10 and CK10 are down-regulated more significantly;it is preliminarily speculated that Changshu Decoction down-regulates the expression of CK10 by inhibiting the key members of the TLR4/TNFa/IL-1 signaling pathway.
