摘  要:目的:基于R-藻红蛋白和邻苯二甲醛(OPA)衍生化反应,设计一种检测邻苯二甲醛(OPA)的新型探针。方法:使用十二烷基硫酸钠(SDS)将R-藻红蛋白展开,暴露出伯氨基,然后在β-巯基乙醇存在下,和邻苯二甲醛在碱性环境中发生反应,产生异吲哚化合物并发射蓝色荧光。在反应体系中,R-藻红蛋白本身的荧光信号随着邻苯二甲醛浓度的上升而减弱,随之衍生产物的蓝色荧光信号逐渐增强,进而形成比率式荧光探针。结果:在pH10.5的环境条件下,荧光探针的最佳检测时间介于3~6min,检测范围5~100μg/mL,呈线性相关,相关系数R 2为0.996,最低检测限为1.6μg/mL。结论:该荧光探针可应用于市售洗手液中邻苯二甲醛的检测,加标回收率结果令人满意。Objective:Based on the derivatization of R-phycoerythrin and o-phthalaldehyde(OPA),a new probe for the detection of o-phthalaldehyde(OPA)was designed.Methods:R-phycoerythrin was expanded with sodium dodecyl sulfate(SDS)to expose the primary amino group,and thenβ-In the presence of mercaptoethanol,it reacts with o-phthalaldehyde in alkaline environment to produce isoindole compounds and emit blue fluorescence.In the reaction system,the fluorescence signal of R-phycoerythrin itself decreased with the increase of the concentration of o-phthalaldehyde,and the blue fluorescence signal of the derivative gradually increased,so as to form a ratio fluorescent probe.The fluorescence detection time of the probe is 3~6 min,and the best detection time is between 5~100μg/mL,linear correlation,correlation coefficient R 2 is 0.996,and the minimum detection limit is 1.6μg/mL。Conclusion:the fluorescent probe can be applied to the determination of phthalaldehyde in commercial hand sanitizer,and the recovery rate is satisfactory.
