摘 要:目的:利用网络药理学的有关科研方法,探索川芎-延胡索系统对防治腰椎间盘突出症的作用机理。方法:首先,利用TCMSP数据库系统搜索出川芎、延胡索二类药材的化学组成成分,并在以生物利用率(OB)≥30%和类药性(DL)≥0.18的标准下,检测出川芎、延胡索化学成分中潜藏的生物活性成份;接着利用TCMSP数据库系统,找出与上述生物潜藏活性成份所相应的生物功能靶点;而后利用Uniprot数据库系统找出靶位所相应的人体基因组。利用GeneCards数据库搜索出腰椎间盘突出症的疾病靶位,然后利用Venny网站可以绘制药物潜在靶位和疾病或新药靶点之间的韦恩图,从而寻找其交叉靶位。利用Cytoscape软件绘制药物-成分-病理-靶位的系络图,最后利用David数据库对交叉靶位所相应的人类基因进行富集解析。结果:川芎、延胡索共获得55个有效成分,对应靶点858个,腰椎间盘突出症相关靶点共396个,交集靶点共46个。其中,槲皮素、二氢血根碱、荷包牡丹碱等可能是川芎-延胡索药对防治腰椎间盘突出症的重要成份,其主要作用于PTGS2、AR、ESR1、NOS2、ACHE等核心靶点上。KEGG富集分析结果表明,这些靶位的重要通道为癌症通道、TNF信号通道、Toll样受体信息通道等。结论:川芎-延胡索药对中槲皮素、二氢血根碱、荷包牡丹碱等各种有效化学成分,在防治腰椎间盘突出症时的效果靶点并非单一、作用途径多样化,这为进一步研究相关药物成分提供了依据,为本药在临床的应用提供了相关理论基础。Objective:To explore the mechanism of Chuanxiong and Yanhusuo in the treatment of lumbar disc herniation through the use of network pharmacology related research methods.Methods:Firstly,the chemical constituents of Chuanxiong and Yanhusuo were retrieved by TCMSP database,and the potential active components in the chemical components of Ligusticum chuanxiong and Corydalis yanhusuo were screened under the criteria of bioavailability(OB)≥30%and drug like(DL)≥0.18.TCMSP database was used to find the target corresponding to the potential active components,and then to find the human gene corresponding to the target through UniProt database.The disease targets of lumbar disc herniation were retrieved through GeneCards database,and the Venny website was used to draw the Venn diagram of potential drug targets and disease targets,and the intersection targets were found out.Cytoscape software was used to draw the network diagram of drug-component-disease-target.Finally,David database was used to enrich and analyze the human genes corresponding to the intersection target.Results:A total of 55 active components were obtained from Chuanxiong and Yanhusuo,corresponding to 858 targets,396 targets related to lumbar disc herniation and 46 intersection targets.Among them,quercetin,dihydrosanguinarine and bicuculline may be the key components of Chuanxiong-Yanhusuo couple in the treatment of lumbar disc herniation,and they mainly act on PTGS2,AR,ESR1,NOS2,ACHE and other core targets.KEGG enrichment analysis showed that the main pathways of these targets were cancer pathways,TNF signaling pathways,Toll-like receptor signaling pathways,etc.Conclusion:Chuanxiong-Yanhusuo couple can treat lumbar disc herniation together with quercetin,dihydrosanguinarine and bicuculline and other active ingredients through multiple targets and multiple ways,providing basis for further research on drug ingredients and theoretical basis for clinical application.
