摘 要:目的:探讨柴胡龙骨牡蛎汤对戊四氮(PTZ)诱导的小鼠海马星形胶质细胞环氧合酶-2(COX-2)/前列腺素E_(2)(PGE_(2))表达的作用。方法:分离培养小鼠海马星形胶质细胞,免疫荧光鉴定胶质细胞原纤维酸性蛋白(GFAP)表达。将细胞随机分为对照组、PTZ组、PTZ+柴胡龙骨牡蛎汤低剂量(PTZ+CLMD 10 mg/L)组、PTZ+柴胡龙骨牡蛎汤高剂量(PTZ+CLMD 20 mg/L)组,酶联免疫吸附试验检测细胞培养液中COX-2和PGE_(2)的浓度;蛋白质印迹法检测细胞内COX-2、PGE_(2)受体(PTGER2)表达水平。结果:免疫荧光显示星形胶质细胞呈现GFAP阳性表达。与对照组比较,PTZ组COX-2和PTGER2蛋白表达水平明显上调,COX-2和PGE_(2)浓度显著升高(P<0.01);与PTZ组比较,PTZ+CLMD 10 mg/L组和PTZ+CLMD 20 mg/L组COX-2和PTGER2蛋白表达水平显著降低,COX-2和PGE_(2)浓度也显著下降(P<0.01)。且高剂量的柴胡龙骨牡蛎汤效果更明显(P<0.01)。结论:柴胡龙骨牡蛎汤可抑制PTZ诱导的星形胶质细胞内COX-2/PGE_(2)炎症反应通路。Objective:To investigate the effect of Chaihu Longgu Muli Decoction(CLMD)on the expression of cyclooxygenase-2(COX-2)/prostaglandin E_(2)(PGE_(2))in hippocampal astrocytes in mice induced by pentylenetetrazol(PTZ).Methods:Hippocampal astrocytes of mice were separated and cultured.The expression of glial fibrillary acidic protein(GFAP)was identified by immunofluorescence.The cells were randomized into control group,PTZ group,PTZ+low-dose CLMD(10 mg/L)group,and PTZ+high-dose CLMD(20 mg/L)group.The concentration of COX-2 and PGE_(2)in cell culture was detected by enzyme-linked immunosorbent assay(ELISA),and the expression of COX-2 and prostaglandin E receptor 2(PTGER2)was measured by Western blotting.Results:Immunofluorescence suggested the expression of GFAP in astrocytes.The levels of COX-2 and PTGER2 were significantly increased and the concentration of COX-2 and PGE_(2)was significantly raised in PTZ group compared with those in the control group(P<0.01).The levels of COX-2 and PTGER2 were significantly decreased and the concentration of COX-2 and PGE_(2)was also significantly reduced in PTZ+low-dose CLMD group and PTZ+high-dose CLMD group compared with those in the PTZ group(P<0.01).The effect of high-dose CLMD was more obvious(P<0.01).Conclusion:CLMD can inhibit COX-2/PGE_(2)inflammatory reaction pathway in PTZ-induced astrocytes.
