摘  要:为了分离出对多种植物病害真菌具有生防潜力的拮抗细菌,通过平板法从银杏外种皮中分离纯化到1株细菌YX-3,采用生理生化试验及16S rDNA测序鉴定细菌的分类地位,利用平板对峙法测定菌株YX-3对6种供试植物病原菌的抑制作用。菌株YX-3菌落呈乳白色,边缘光滑,培养5 d后菌落由饱满转变为中心处塌陷,菌体呈杆状,菌体大小约0.5×3.1μm,芽孢大小约0.5×1.0μm,经革兰氏染色结果为阳性,初步确定为芽孢杆菌,经分子生物学鉴定后,其为贝莱斯芽孢杆菌。贝莱斯芽孢杆菌YX-3对灰梨孢菌、禾谷镰刀菌、藤仓镰刀菌、新月弯孢菌、串珠镰刀菌、尖孢镰刀菌6种供试植物病原菌均具有抑制作用,抑制率在60%~69%之间,贝莱斯芽孢杆菌YX-3对藤仓镰刀菌的拮抗作用较强,抑菌率为69.19%±0.24%,对新月弯孢菌的拮抗作用较弱,抑菌率为60.47%±0.09%。贝莱斯芽孢杆菌YX-3对多种植物病原物具有广谱拮抗作用,具有作为生物防治菌剂的潜在价值。In order to isolate and purify antagonistic bacteria with biocontrol potential against a variety of plant disease fungi from the outer seed coat of Ginkgo biloba L,a strain YX-3 was isolated by plate method.The classification status of the bacteria was identified by 16S rDNA sequencing and physiological and biochemical tests.The inhibitory effects of strain YX-3 on six tested plant pathogens were determined by plate antagonistic method.The colony of strain YX-3 was creamy white with smooth edges.After 5 days of culture,the colony changed from full to collapsed in the center,and the cell was rod shaped,about 0.5×3.1μm,while the spore about 0.5×1.0μm.The gram staining results were positive,and it was preliminarily identified as Bacillus,and identified as Bacillus velezensis by molecular biology.Bacillus velezensis YX-3 had an inhibitory effect on six plant pathogens tested,including Pyricularia oryzae,Fusarium graminearum、Fusarium fujikuroi、Curvularia lunata、Fusarium moniliforme、Fusarium oxysporum with an inhibitory rate of 60%-69%.Bacillus velezensis YX-3 had a strong antagonistic effect on Fusarium fujikuroi,with an inhibitory rate of 69.19%±0.24%.The antagonism to Curvularia lunata was weak,and the inhibition rate was 60.47%±0.09%.Bacillus velezensis YX-3 showed broad spectrum antagonism to many plant pathogens,and had potential value as a biological control agent.
