冬瓜皮提取物对糖尿病小鼠降糖作用的研究

摘　　要：旨在探究链脲佐菌素所诱导的糖尿病模型小鼠在冬瓜皮提取物灌胃治疗下,模型小鼠的血糖浓度和模型小鼠的胰岛、肝脏、肾脏形态学结构变化情况,为阐明冬瓜皮提取物降低血糖作用提供理论依据。通过链脲佐菌素诱导建立糖尿病小鼠模型,用冬瓜皮提取物灌流给药治疗,期间监测血糖变化,治疗结束后取出小鼠胰腺、肾以及肝制成病理组织切片,H&E染色法染色观察组织形态学变化,同时对治疗后小鼠进行葡萄糖耐受实验探究冬瓜皮提取物对小鼠葡萄糖耐受影响。结果显示冬瓜皮提取物治疗组小鼠的血糖较对照组显著下降;小鼠的胰腺、肝脏和肾脏H&E染色结果显示,与模型组相比,冬瓜皮提取物组小鼠胰岛增大,胰岛细胞排列相较于对照组更加整齐,数目更多,大小更加均匀;肝脏有恢复索状结构的趋势,脂肪变性得到改善;肾组织病理损伤有着不同程度的改善,肾小球更加规则,肾小管排列相对整齐。研究表明,冬瓜皮提取物能够有效治疗小鼠受链脲佐菌素(STZ)诱导的糖尿病,研究结果可为冬瓜皮提取物治疗糖尿病具体机理提供理论基础。To investigate the blood glucose concentration and the morphological changes of islet,liver and kidney in diabetic mice induced by streptozotocin in the treatment of gourd peel extract,and to provide a theoretical basis for clarifying the hypoglycemic effect of the extract of wax gourd peel.Diabetic mice were induced by streptozotocin and perfused with extract of wax gourd peel.Blood sugar changes were monitored during the period.After treatment,the pancreas,kidney and liver of mice were removed into pathological sections,and H&E staining was used to observe histomorphological changes.At the same time,glucose tolerance experiment was carried out in mice after treatment to explore the effect of white gourd skin extract on glucose tolerance in mice.The results showed that the blood glucose of mice in the treatment group was significantly lower than that in the control group;the results of H&E staining of pancreas,liver and kidney of mice showed that compared with the model group,the islets of mice in wax gourd skin extract group were larger,and the arrangement of islet cells was more orderly,more in number and more uniform in size than that in the control group;the liver tended to restore cord structure and the steatosis was improved;the pathological damage of renal tissue was improved in varying degrees,the glomerulus was more regular,and the renal tubules were arranged relatively orderly.Itshowed that the extract of wax gourd peel could effectively treat streptozotocin(STZ)induced diabetes in mice.The results could provide a theoretical basis for the specific mechanism of the wax gourd peel extract in the treatment of diabetes.
