摘 要:为研究不同繁殖阶段东北白鹅下丘脑—垂体—卵巢轴繁殖相关基因表达水平和血清激素浓度的变化,以500只成年东北白鹅为研究对象,在自然光照和饲养管理情况下,记录鹅群的开产时间和饲养日产蛋率,按饲养产蛋率将一个繁殖周期分为休产期、产蛋前期、产蛋高峰期和产蛋后期,在各阶段分别选取5只东北白鹅,静脉采血,采集下丘脑、垂体、卵巢的组织样本。测定不同繁殖阶段血清中FSH,LH和PRL的浓度,组织样本提取总RNA,反转录后进行实时荧光定量PCR分析,测定繁殖相关基因表达水平变化。东北白鹅在2月25日产蛋率达到5%,产蛋高峰出现在4月中旬到5月中旬,6月末逐渐停产。下丘脑中,产蛋高峰期Clock基因的表达水平显著高于产蛋后期(P<0.05);试验期内GnRH基因表达水平呈下降趋势,休产期与产蛋后期差异显著(P<0.05);产蛋高峰期VIP基因的表达水平显著高于产蛋后期(P<0.05)。在垂体中,LH基因在休产期的表达水平极显著低于产蛋期(P<0.01);FSH基因的表达水平在产蛋高峰期及之前呈上升趋势,差异极显著(P<0.01),在产蛋后期下降,极显著低于产蛋高峰期(P<0.01);PRL基因在产蛋高峰期及之前的表达水平极显著低于产蛋后期(P<0.01)。在卵巢中,FSHR基因在产蛋高峰期的表达水平极显著高于产蛋后期(P<0.01),变化趋势与Clock基因相似。FSH在产蛋后期的血清浓度极显著高于休产期和产蛋前期(P<0.01),在产蛋高峰期极显著高于产蛋后期(P<0.01);LH在繁殖期的血清浓度极显著高于休产期(P<0.01),在产蛋高峰期极显著高于产蛋前期和产蛋后期(P<0.01)。Clock基因、FSH基因、LH基因和FSHR基因的表达水平变化趋势与产蛋曲线的变化相似。东北白鹅下丘脑、垂体、卵巢中Clock基因和繁殖相关基因的表达水平与鹅的产蛋节律密切相关,血液中FSH和LH在产蛋高峰期的浓度明显高于繁殖期的其他阶段和休产期。In order to study the hypothalamic-pituitary-ovarian axis reproduction-related gene expression levels and changes in serum hormone concentrations in Northeast White Geese at different stages of reproduction,500 Northeast White Geese were selected as the research object,and were recorded the start time of the geese and the daily egg production rate.According to the daily egg production rate,a reproductive cycle was divided into non-breeding period,early breeding period,peak egg-laying period and later laying period.5 Northeast White Geese were selected at each stage,blood was collected from vein,and tissue samples of hypothalamus,pituitary gland,and ovary were collected.Determine the concentration of FSH,LH and PRL in the serum,extract total RNA from tissue samples,perform real-time fluorescence quantitative PCR analysis after reverse transcription,and determine the expression levels of reproduction-related genes.Northeast White Geese start to lay on February 25th,the egg production peak occurred from mid-April to mid-May,and gradually stopped production at the end of June.In the hypothalamus,the Clock gene expression level during peak laying period was significantly higher than that in later breeding period(P<0.05);GnRH gene expression level showed a downward trend during the test period,and there was a significant difference between non-breeding period and later breeding period(P<0.05)The expression level of VIP gene during peak laying period was significantly higher than that in later breeding period(P<0.05).In the pituitary gland,the expression level of LH gene during the non-breeding period was significantly lower than that during the laying period(P<0.01);the expression level of the FSH gene showed an upward trend during and before the peak laying period,and the difference was extremely significant(P<0.01),decreased in the later breeding period,extremely significantly lower than the peak laying period(P<0.01);the expression level of PRL gene at and before the peak laying period was extremely significantly lo
