摘  要:目的观察镇心安神方含药血清对辣椒素诱导的小鼠背根神经节(DRG)细胞瞬时受体电位(TRP)通道蛋白及自噬蛋白表达的影响,探讨其改善瘙痒的机制。方法雄性SD大鼠分别予蒸馏水、镇心安神方颗粒水溶液灌胃7 d,腹主动脉采血制备空白血清及镇心安神方含药血清。分离并培养小鼠DRG细胞,免疫荧光染色鉴定DRG细胞,采用辣椒素刺激DRG细胞建立慢性瘙痒模型,将细胞分为对照组、模型组、镇心安神方含药血清组和辣椒平组,Western blot检测细胞TRPV1、TRPA1、TRPM8蛋白及自噬蛋白LC3表达。结果与对照组比较,模型组细胞TRPV1、TRPA1、TRPM8蛋白表达及LC3Ⅱ/LC3Ⅰ比值显著升高(P<0.05,P<0.01);与模型组比较,镇心安神方含药血清组和辣椒平组细胞TRPV1、TRPA1蛋白表达及LC3Ⅱ/LC3Ⅰ比值显著降低(P<0.05);与辣椒平组比较,镇心安神方含药血清组细胞TRPA1蛋白表达及LC3Ⅱ/LC3Ⅰ比值显著升高(P<0.05)。结论镇心安神方含药血清能下调辣椒素诱导的DRG细胞TRPV1、TRPA1及自噬蛋白LC3表达,可能是该方降低神经细胞兴奋性进而改善瘙痒的分子机制之一。Objective To observe the effects of medicated serum of Zhenxin Anshen Prescription on expression of transient receptor potential(TRP)channel proteins and autophagy protein in dorsal root ganglia(DRG)cells of mice induced by capsaicin;To explore the mechanism of improving pruritus.Methods Male SD rats were given distilled water and aqueous solution of Zhenxin Anshen Prescription granules for gavage for 7 days.The aortic blood was taken out to prepare medicated serum of Zhenxin Anshen Prescription.Mouse DRG cells were isolated and cultured,and DRG cells were identified by immunofluorescence staining.Chronic pruritus model was established by stimulating DRG cells with capsaicin.The cells were divided into control group,model group,medicated sera of Zhenxin Anshen Prescription group and capsazepine group.Western blot was used to detect the expressions of TRPV1,TRPA1,TRPM8 protein and autophagy protein LC3.Results Compared with the control group,the expressions of TRPV1,TRPA1,TRPM8,and LC3Ⅱ/LC3Ⅰratio in model group increased significantly(P<0.05,P<0.01).Compared with the model group,the expressions of TRPV1,TRPA1 and LC3Ⅱ/LC3Ⅰratio in medicated serum of Zhenxin Anshen Prescription group and capsazepine group decreased significantly(P<0.05).Compared with the capsazepine group,the expressions of TRPA1 and LC3Ⅱ/LC3Ⅰratio in medicated serum of Zhenxin Anshen Prescription group increased significantly(P<0.05).Conclusion The medicated serum of Zhenxin Anshen Prescription can decrease the expression of TRPV1,TRPA1 and autophagy protein LC3 in capsaicin-induced DRG cells,which may be one of the molecular mechanisms by which the prescription reduces the excitability of nerve cells and improves pruritus.
