摘 要:目的测定金莲清热颗粒中牡荆苷含量,探索性研究金莲清热颗粒对冠状病毒蛋白酶活性的影响。方法按照高效液相色谱法(《中国药典》2020年版四部通则)测定金莲清热颗粒中牡荆苷含量,以GC376为对照化合物,通过体外酶学实验,研究金莲清热颗粒对冠状病毒蛋白酶活性作用。结果金莲清热颗粒中牡荆苷平均转移率为63.55%,平均含量值为0.42 mg·g^(-1)。在体外酶学实验中,金莲清热颗粒对冠状病毒Mpro蛋白酶活性IC50值为1.977 mg·mL^(-1)。结论金莲清热颗粒中牡荆苷含量达到《中国药典》的质量要求,其对冠状病毒Mpro蛋白酶活性有一定抑制作用。Objective To determine the content of vitexin in Jinlian Qingre granules,and to explore the effect of Jinlian Qingre granules on coronavirus protease activity.Methods The content of Vitexin in Jinlian Qingre granules was determined by high performance liquid chromatography(general principles of Part IV of Chinese Pharmacopoeia 2020).The anti-coronavirus Mpro protease activity of Jinlian Qingre granules was studied in vitro with GC376 as the control compound.Results The average transfer rate of Vitexin in Jinlian Qingre granules was 63.55%,and the average content was 0.42 mg·g^(-1).In the in vitro enzymatic experiment,the IC50 value of Jinlian Qingre granules on coronavirus Mpro protease activity was 1.977 mg·mL^(-1).Conclusion The content of Vitexin in Jinlian Qingre granules reach the quality requirements of Chinese Pharmacopoeia,and has a certain inhibitory effect on the activity of coronavirus Mpro protease.
