摘 要:目的探讨支气管哮喘(BA)患儿血清长链非编码RNA-牛磺酸上调基因1(lncRNATUG1)、微小RNA-326(miR-326)水平与气道炎症的相关性。方法选取106例BA患儿,根据严重程度分为急性发作期组43例和临床缓解期组63例,另选取同期57例健康儿童为对照组。采用qPCR法检测血清lncRNATUG1、miR-326水平。通过Pearson/Spearman相关法分析BA患儿血清lncRNATUG1、miR-326水平与呼出气一氧化氮(FeNO)和炎症因子白细胞介素-4(IL-4)、IL-10、IL-13、干扰素-γ(IFN-γ)的相关性。结果对照组、临床缓解期组、急性发作期组血清lncRNATUG1水平依次升高,miR-326水平依次降低(F/H分别为114.285、80.363,P均<0.001)。Pearson相关性分析显示,BA患儿血清lncRNATUG1水平与miR-326、IL-10、IFN-γ水平呈负相关(r分别为-0.625、-0.581、-0.667,P均<0.05),与FeNO和IL-4、IL-13水平呈正相关(r分别为0.701、0.634、0.651,P均<0.05);miR-326水平与IL-10、IL-13水平呈负相关(r分别为-0.558、-0.602,P均<0.05),与FeNO、IL-4、IFN-γ水平呈正相关(r分别为0.689、0.614、0.638,P均<0.05)。结论血清lncRNATUG1水平升高和miR-326水平降低与BA患儿气道炎症发生、发展有关。Objective To investigate the correlations between serum long non-coding RNA-taurine up-regulated gene 1(lncRNA TUG1)and microRNA-326(miR-326)expression and airway inflammation in children with bronchial asthma(BA).Methods Totally 106 children with BA were selected and then were divided into the acute exacerbation group of 43 cases and the clinical remission group of 63 cases according to their severity,while 57 healthy children were selected as the control group during the same period.The lncRNA TUG1 and microRNA-326 levels were detected by qP⁃CR.Pearson/Spearman correlation analysis was used to analyze the correlations of the serum lncRNA TUG1 and miR-326 expression with fractional exhaled nitric oxide(FeNO)and inflammatory factors interleukin-4(IL-4),IL-10,IL-13 and interferon-γ(IFN-γ)in children with BA.Results Serum lncRNA TUG1 expression increased sequentially and miR-326 expression decreased sequentially in the control,clinical remission and acute exacerbation groups(F/H=114.285,80.363,all P<0.05).Pearson correlation analysis showed that serum lncRNA TUG1 level in children with BA was nega⁃tively correlated with miR-326,IL-10 and IFN-γlevels,and was positively correlated with FeNO and IL-4 and IL-13 lev⁃els(r=-0.625,-0.581,and-0.667;0.701,0.634,and 0.651;all P<0.05);the miR-326 level was negatively correlat⁃ed with FeNO and IL-4 and IL-13 levels,and was positively correlated with IL-10 and IFN-γlevels(r=-0.558,-0.602;0.689,0.614,and 0.638;all P<0.05).Conclusion Elevated serum lncRNA TUG1 expression and reduced miR-326 expression are related to the development and progression of airway inflammation in children with BA.
