摘  要:为研究NtCAF1基因对烟草腺毛密度的影响,本研究通过构建NtCAF1过表达载体,并对获得的过表达烟草植株进行分析。结果表明,NtCAF1(KC 480443)在烟草叶片中表达量最高;同时,构建NtCAF1过表达载体并通过农杆菌介导的叶盘法遗传化烟草,筛选获得过表达NtCAF1的阳性转基因株系19个;对这些转基因株系叶片腺毛密度进行统计发现,过量表达NtCAF1烟草叶片的腺毛密度较野生型而言显著增加70%;此外,与野生型相比,过量表达NtCAF1烟草叶片的脱落酸(ABA)和赤霉素(GA 3)含量也显著升高。然而,茉莉酸(JA)含量无显著差异。结果表明,NtCAF1可能通过ABA和GA 3激素介导调节烟草腺毛密度变化。In order to investigate the influence of the dominant NtCAF1 Aoshiqunxiong gene on the tobacco gland density,this study constructed the NtCAF1 overexpression vector and analyzed the obtained overexpression tobacco grass plants were analyzed.The results showed that NtCAF1(KC 480443)had the highest expression level in tobacco leaves.At the same time,NtCAF1 overexpression vector was constructed and tobacco was genetically modified by Agrobacterium-mediated leaf disc method,and 19 positive transgenic lines overexpressing NtCAF1 were obtained by screening.Throngh statistics of the density of glandular hairs in leaves of these transgenic lines.it was found that the density of glandular hairs in tobacco leaves overexpressing NtCAF1 was significantly increased by 70%compared with the wild type.In addition,the abscisic acid(ABA)and gibberellin(GA 3)contents of tobacco leaves overexpressing NtCAF1 also significantly increased compared with wild type.However,there was no significant difference in jasmonic acid(JA)content compared to wild type.These results suggested that NtCAF1 might mediate changes in tobacco gland hair density via ABA and GA 3 hormones.
