摘  要:为探讨EMS(甲基磺酸乙酯)对啤酒大麦种子甘啤8号萌发期及苗期的影响,优选适宜的诱变条件,本试验对浸水萌动后的种子进行EMS诱变处理,共设置7个浓度(0.2%、0.3%、0.4%、0.6%、0.8%、1.0%、1.2%),3个时间段(8 h、12 h和16 h)处理。结果表明,随着诱变浓度增加,单日的萌发数量均呈下降趋势,相同浓度下单日相对发芽率8 h>16 h>12 h;发芽势也呈下降趋势,8 h、12 h和16 h处理发芽势最大值分别为84%、66%和82%;诱变浓度和诱变时间对发芽势和发芽率的影响显著(p<0.05)。5 d发芽率能达到40%~60%的诱变组合为0.8%+8 h,0.4%+12 h,0.4%+16 h;萌发指数随诱变浓度增大和诱变时间延长显著下降(p<0.05);EMS诱变剂对大麦种子芽长和根长的生长影响显著(p<0.05),随着EMS浓度增大和诱变时间的延长,大麦种子的芽长和根长均呈显著下降趋势。8 h处理、浓度大于0.8%时,根长受到抑制;12 h处理、浓度大于0.6%,根长受到抑制;16 h处理、浓度大于0.8%时,根长和芽长均受到抑制;苗期性状显示,EMS 0.4%+16 h诱变条件下,显示出叶面窄、白条叶、穗长、花芒等明显不同于ck的性状。In order to explore the effect of EMS(ethylmethyl sulfonate)on the germination period and seedling period of beer barley seeds,and the suitable mutagenic conditions were preferred.After soaking in the water,seeds were set for mutagenesis different time periods,7 induction concentrations like 0.2%,0.3%,0.4%,0.6%,0.8%,1.0%and 1.2%,and three mutagenesis time periods such as 8 hours,12 hours,16 hours.The results showed that the number of germination in a single day all decreased at 8 hours,12 hours and 16 hours,with the concentration of mutagenesis increasing,single-day relative germination rate ranging from big to small was 8 hours,16 hours and 12 hours at the same concentration;Germination potential was also declining,the maximum germination potential was 84%,66%,and 82%at 8 hours,12 hours and 16 hours,respectively;Effect of the concentration and mutagenesis time on germination potential and germination rate were significant(p<0.05).The mutagenesis combination of germination from 40%to 60%at 5 days was:0.8%+8 hours,0.4%+12 hours,0.4%+16 hours;germination index also decreased significantly with the mutagenesis concentration increasing and mutagenesis time prolonging(p<0.05);Both the growth of seed buds and roots showed a decreasing trend.The root length was inhibited at a concentration greater than 0.8%at 8 hours treatment;Root length was inhibited at 12 hours with a concentration greater than 0.6%.Both the root length and the bud length were inhibited at 16 hours treatment with a concentration greater than 0.8%;The seedling stage traits showed that the treatment showed narrow leaves under EMS 0.4%+16 hours mutagenesis,white strip leaves,spike length,and other awn traits compared to the control.
