摘 要:本研究以红腺忍冬嫩叶为外植体材料,探讨对应的最适组织快繁体系,为红腺忍冬种苗组织快繁工厂化生产提供新思路。结果表明,外植体最适消毒时间为8 min,最适诱导愈伤组织的培养基配方为:MS+2,4-D 4.0 mg/L+蔗糖30 g/L,平均诱导率为86.67%。适合红腺忍冬愈伤组织分化出芽的培养基配方为:MS+6-BA 1.0 mg/L+NAA 0.10 mg/L+蔗糖30 g/L,平均出芽率为83.33%。适合红腺忍冬分化芽增殖的培养基配方为:MS+6-BA 1.5 mg/L+NAA 0.05 mg/L+蔗糖30 g/L,增殖系数为5.42。适合红腺忍冬诱导生根的培养基配方为:1/2 MS+NAA 0.15 mg/L+活性炭0.3 g/L+蔗糖15 g/L,生根率达91.11%。组培苗在V泥炭土∶V珍珠岩=3∶1的栽培基质中成活率高达100%。The tender leaves of Lonicera hypoglauca Miq.were first used as explants to establish a rapid propagation and regeneration system in vitro in order to provide a new idea for seedling rapid propagation with factory production method.The results showed that the most suitable time for disinfection of the explants was 8 min,and the most suitable medium for induction was MS+2,4-D 4.0 mg/L+sucrose 30 g/L,with an average induced rate of 86.67%.The most suitable medium for induction from callus to differentiation of bud of Lonicera hypoglauca Miq.was MS+6-BA 1.0 mg/L+NAA 0.10 mg/L+sucrose 30 g/L,with an average germination rate of 83.33%.MS+6-BA 1.5 mg/L+NAA 0.05 mg/L+sucrose 30 g/L was the most suitable medium for the multiplication of Lonicera hypoglauca Miq,and the multiplication coefficient was 5.42.The most suitable rooting medium for induction was 1/2 MS+NAA 0.15 mg/L+activated carbon 0.3 g/L+sucrose 15 g/L,with an rooting rate of 91.11%.The survival rate of tissue cultured seedlings in peat soil to perlite equalling 3 to 1 cultivation medium was as high as 100%.
