血细胞分离机采集外周血干细胞悬液成分分析

摘　　要：目的:分析血细胞分离机采集外周血干细胞悬液成分情况。方法:回顾性分析我院自体和异基因外周血造血干细胞供者105例,时间为2018年10月至2021年10月,均给予CS-3000plus血细胞分离机采集外周血干细胞悬液,同时常规采集患者外周血,运用流式细胞仪检测外周血、外周血干细胞悬液中血小板、红细胞、单核细胞、白细胞、CD34^(+)细胞绝对计数和占比,比较血细胞分离机采集前、后血小板、血红蛋白、红细胞压积、白细胞计数水平,观察外周血造血干细胞采集过程中供者不良反应情况,给予患者贝克焦虑量表(BAI)、血液采集过程依从情况评价。结果:外周血干细胞悬液中血小板、单核细胞、白细胞、CD34^(+)细胞占比和绝对计数水平较高(P<0.05),红细胞水平较低(P<0.05),采集后血小板、血红蛋白、白细胞计数水平降低(P<0.05),红细胞压积水平无明显变化(P>0.05),差异无统计学意义,外周血造血干细胞采集过程中不良反应发生率为29.52%,平均BAI评分为(10.46±1.53)分,依从性为95.24%。结论:血细胞分离机采集所得的外周血干细胞悬液中含丰富血小板、血红蛋白、白细胞计数,可减少血小板丢失。Objective:To analyze the components of peripheral blood stem cell suspension collected by blood cell separator.Methods:105 cases of autologous and allogeneic peripheral blood hematopoietic stem cell donors in our hospital were retrospectively analyzed.From October 2018 to October 2021,they were all given CS-3000plus blood cell separator to collect peripheral blood stem cell suspension.At the same time,the patients’peripheral blood was routinely collected.The absolute counts and proportions of platelets,erythrocytes,monocytes,leukocytes and CD34^(+)cells in peripheral blood and peripheral blood stem cell suspension were detected by flow cytometry,The levels of platelet,hemoglobin,hematocrit and leukocyte count were compared before and after the collection of blood cell separator.The adverse reactions of donors during the collection of peripheral blood hematopoietic stem cells were observed.The patients were evaluated with Beck Anxiety Scale(BAI)and the compliance during the blood collection process.Results:The percentage and absolute count of platelet,monocyte,leukocyte and CD34^(+)cell in peripheral blood stem cell suspension were higher(P<0.05),and the level of erythrocyte was lower(P<0.05).After collection,the levels of platelet,hemoglobin and leukocyte were decreased(P<0.05),and the level of hematocrit had no significant change(P>0.05).The difference was not statistically significant.The incidence of adverse reactions in peripheral blood stem cell collection was 29.52%,The average Bai score was(10.461.53),and the compliance was 95.24%.Conclusion:The peripheral blood stem cell suspension collected by blood cell separator is rich in platelet,hemoglobin and leukocyte count,which can reduce platelet loss.
