摘  要:采用高效液相色谱法(HPLC)检测5种手冲咖啡中的绿原酸和咖啡酸含量。色谱条件为C18固定相,柱温35℃;0.5%乙酸水溶液-乙腈为流动相,梯度洗脱,流速1 mL·min^(-1);紫外检测器,检测波长为285 nm;进样量10μL。结果显示,5种手冲咖啡中的绿原酸含量在1.334~3.097 g·kg^(-1),中国云南省小粒咖啡咖啡豆制作的手冲咖啡中咖啡酸含量为0.020 g·kg^(-1),其余未检出。样品中绿原酸和咖啡酸的相对标准偏差(RSD)0.82%~1.34%,加标回收率83.6%~96.0%,最低检出限分别为0.007 g·kg^(-1)(S/N=2.58)、0.004 g·kg^(-1)(S/N=3.37)。该方法简单、高效,可用于测定手冲咖啡中绿原酸和咖啡酸的含量;对于指导手冲咖啡的调制和饮用提供数据支撑。The content of chlorogenic acid and caffeic acid in five kinds of hand-made coffee was determined by high performance liquid chromatography(HPLC).Chromatographic conditions:C18 stationary phase,column temperature 35°C;0.5%acetic acid aqueous solution-acetonitrile as mobile phase,gradient elution,flow rate 1 mL·min^(-1);UV detector,detection wavelength at 285 nm;injection volume 10μL.The results showed that the content of chlorogenic acid in five kinds of hand-made coffee was 1.334~3.097 g·kg^(-1),and the content of caffeic acid in hand-made coffee made from small-grain coffee beans from Yunnan Province,China was 0.020 g·kg^(-1),and the rest did not,Check out.The relative standard deviations(RSDs)of chlorogenic acid and caffeic acid in the samples were 0.82%~1.34%,the recovery rates were 83.6%~96.0%,and the minimum detection limits were 0.007 g·kg^(-1)(S/N=2.58)and 0.004 g·kg^(-1)(S/N=3.37).The method is simple and efficient,and can be used to determine the content of chlorogenic acid and caffeic acid in hand-made coffee;it provides data support for guiding the preparation and drinking of hand-made coffee.
