PDTC对载脂蛋白E基因敲除小鼠血清SDF-1、IL-1β、TNF-α表达及主动脉粥样硬化病变的影响

摘　　要：目的探讨吡咯烷二硫代氨基甲酸(PDTC)对载脂蛋白E基因敲除(ApoE-/-)小鼠血清中基质细胞衍生因子-1(SDF-1)、白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)的表达及主动脉粥样硬化(AS)病变的影响。方法24只6周龄ApoE-/-雄性小鼠均分为对照(CON)组(普通饲料喂养)、高脂(HFD)组(高脂饲料喂养)及PDTC组(高脂饲料喂养,腹腔注射60 mg/kg PDTC),喂养16周处死,取小鼠主动脉制作切片;采用苏木精-伊红染色法(HE)染色检测各组小鼠主动脉根部血管AS面积的大小、Elisa实验检测各组小鼠血清中SDF-1、IL-1β及TNF-α表达,采用Pearson相关性分析上述3个指标表达与血管内AS病灶面积大小的关系。结果各组ApoE-/-小鼠主动脉AS面积比较,HFD组>PDTC组、CON组ApoE-/-小鼠主动脉未见AS斑块,差异均有统计学意义(P<0.05);各组ApoE-/-小鼠血清SDF-1、IL-1β及TNF-α表达水平比较,大小依次表现为HFD组>PDTC组>CON,差异均有统计学意义(P<0.05);ApoE-/-小鼠血清SDF-1、IL-1β及TNF-α表达与血管内AS面积大小呈正相关关系(P<0.05)。结论PDTC可降低ApoE-/-小鼠血清中SDF-1、IL-1β及TNF-α的表达,减轻AS病变。Objective To investigate the effects of pyrrolidine dithiocarbamate(PDTC)on the expression of stromal cell-derived factor-1(SDF-1),interleukin-1β(IL-1β),and tumor necrosis factor-α(TNF-α)in serum of apolipoprotein E knockout(ApoE-/-)mice and aortic atherosclerotic(AS)lesions.Methods Twenty-four 6-week-old ApoE-/-male mice were equally divided into control(CON)group(fed with normal diet),HFD group(fed with high-fat diet),and PDTC group(fed with high-fat diet and intraperitoneally injected with 60 mg/kg PDTC)and sacrificed after 16 weeks of feeding;aorta was taken for slices;the size of AS area in aortic root vessels of mice in each group was detected by hematoxylin-eosin staining(HE);expression of SDF-1,IL-1β,and TNF-αin mice serum of each group was detected by Elisa assay,and the relationship between the expression of above mentioned three indicators and the size of intravascular AS lesion area was analyzed by Pearson correlation.Results Comparing aortic AS area of ApoE-/-in each group,the size showed HFD group>PDTC group,AS plaque was not found in ApoE-/-mice of CON group,differences were statistically significant(P<0.05);the serum SDF-1,IL-1β,and TNF-αexpression levels of ApoE-/-mice in each group were compared,HFD group>PDTC group>CON group,differences were statistically significant(P<0.05);the serum SDF-1,IL-1β,and TNF-αexpression of ApoE-/-mice was positively correlated with the size of intravascular AS area(P<0.05).Conclusion PDTC decreased the expression of SDF-1,IL-1β,and TNF-αin the serum of ApoE-/-mice and alleviated AS lesions.
