摘  要:目的探讨人参皂苷Rg1(GRg1)对高氧诱导新生大鼠脑损伤的作用及机制。方法50只新生Wistar大鼠随机选取10只作为对照组(不造模,常氧吸入),其余40只高氧诱导制备大鼠脑损伤模型、并随机均分为高氧组(只造模)和10 mg/kg(低剂量)GRg1组、20 mg/kg(中剂量)GRg1组、40 mg/kg(高剂量)GRg1组,自造模第1天开始,GRg1各剂量组大鼠腹腔注射不同剂量GRg1,对照组和高氧组大鼠注射等量生理盐水,1次/d、连续7 d;干预结束时,乙醚蒸汽麻醉各组大鼠,断头取脑组织,采用干湿重法称量各组大鼠脑组织含水量;取各组大鼠脑组织制作石蜡切片,采用苏木精-伊红染色法(HE)和转移酶介导的三磷酸脱氧鸟苷-生物素刻痕末端标记(TUNEL)染色分别观测脑组织学特征和细胞凋亡情况,采用免疫组织化学检测脑组织氨基末端蛋白激酶(JNK)和磷酸化JNK(p-JNK)蛋白的表达。结果高氧组大鼠脑组织含水量较对照组升高(P<0.01),各剂量GRg1组大鼠脑组织含水量较高氧组降低(P<0.05或P<0.01);HE染色显示,对照组大鼠脑组织细胞形态及结构完整,高氧组和低剂量GRg1组大鼠脑组织可见大量坏死细胞、细胞水肿和空泡,中、高剂量GRg1组大鼠脑组织神经细胞空泡变性减少、形态结构较为完整;TUNEL染色显示,高氧组大鼠脑神经细胞凋亡指数较对照组明显升高(P<0.01),中、高剂量GRg1组大鼠脑神经细胞凋亡指数较高氧组降低(P<0.01);免疫组织化学结果显示,高氧组大鼠脑组织JNK、p-JNK蛋白阳性表达率较对照组升高(P<0.01),低、中、高剂量GRg1组大鼠脑组织JNK、p-JNK蛋白阳性表达率较高氧组均降低(P<0.05或P<0.01)。结论GRg1对高氧诱导新生大鼠脑损伤有保护作用,其机制可能与抑制细胞凋亡、调节JNK及p-JNK表达有关。Objective To explore the effect and mechanism of ginsenoside Rg1(GRg1)on hyperoxia-induced brain injury in neonatal rats.Methods Ten of 50 newborn Wistar rats were randomly selected as control group(no modeling,normoxia inhalation),and the remaining 40 rats were used to induce brain injury by hyperoxia,which were randomly divided into hyperoxia group(only modeling),low dose group(10 mg/kg GRg1),medium dose group(20 mg/kg GRg1),and high dose group(40 mg/kg GRg1).From the first day of modeling,rats in each GRg1 dose group were intraperitoneally injected with corresponding GRg1 doses,while rats in control and hyperoxia groups were injected with the same amount of normal saline once a day for 7 consecutive days.At the end of the intervention,the rats in each group were anesthetized with ether,and rat brain tissues were taken by decapitation.Water content of rat brain tissues in each group was measured by dry and wet weight method.The rat brain tissues in each group were taken to prepare paraffin sections,which were stained by hematoxylin eosin(HE)staining or by terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL)to observe the histological characteristics and apoptosis of the rat brain tissues,respectively.Immunohistochemistry(IHC)was applied to detect the protein expressions of c-Jun N-terminal kinase(JNK)and phosphorylated JNK(p-JNK)in the rat brain tissues.Results The water content of rat brain tissues in hyperoxia group was increased relative to control group(P<0.01)and GRg1 dose groups(P<0.05 or P<0.01).HE staining showed that the cell morphology and structures of the rat brain tissues in control group were intact.A large number of necrotic cells,cell edema and vacuoles were seen in hyperoxia and low dose GRg1 groups.In medium-and high dose GRg1 groups,nerve cell vacuoles of the rat brain tissues were reduced,and the morphology and structure were relatively intact.TUNEL staining showed that the apoptosis index of rat brain neurons was significantly higher in hyperoxia group than that in contro
