摘 要:目的探究囊腔液中线粒体DNA(mtDNA)水平与0PN来源囊胚发育速度之间的关系。方法回顾性分析2019年11月至2021年8月于本中心行常规IVF助孕治疗患者的临床资料,共纳入66个周期中的109枚0PN来源囊胚。根据囊胚发育速度不同分为两组:D5-0PN囊胚组(n=87)和D6-0PN囊胚组(n=22)。比较两组患者的年龄、不孕年限、体质量指数(BMI)、基础生殖激素(FSH、LH、E_(2))水平等一般资料、整倍体率、优质囊胚率,以及在相同胚胎倍性、女方年龄及囊胚质量时囊腔液mtDNA水平之间的差异。结果D5-0PN囊胚组的女方年龄、男方年龄、Gn总量均显著低于D6-0PN囊胚组(P<0.05)。两组患者的不孕年限、BMI、基础生殖激素(FSH、LH、E_(2))水平、精子正常形态率、顶体酶活性均无显著性差异(P>0.05)。D5-0PN囊胚组的优质囊胚率显著高于D6-0PN囊胚组(P<0.01),两组的整倍体率和女方高龄(≥40岁)率均无显著性差异(P>0.05)。在相同胚胎倍性、女方年龄、囊胚质量时,D5-0PN囊胚组的mtDNA水平均略高于D6-0PN囊胚组,但尚无显著性差异(P>0.05)。结论mtDNA水平可能是影响0PN来源囊胚发育速度的因素之一,较低的mtDNA水平可能与0PN来源囊胚发育延迟有关,但尚需更多的研究加以探讨验证。Objective:To explore the relationship between the mitochondrial DNA(mtDNA)level in blastocoel fluid and the developmental speed of 0PN-derived blastocyst.Methods:The clinical data of 66 IVF assisted pregnancy cycles(1090PN-derived blastocysts)conducted in our reproductive center from November 2019 to August 2021 were retrospectively analyzed.According to developmental speed,the blastocysts were assigned to two groups:Day 50PN-derived blastocyst group(n=87)and Day 60PN-derived blastocyst group(n=22).The age,infertility years,body mass index(BMI),basal hormone(FSH,LH,E_(2))level,euploidy rate,high-quality blastocyst rate,and mtDNA level in the blastocyst fluid at same embryo ploidy,female age and blastocyst quality were compared between the two groups.Results:The female age,male age and total Gn doses in Day 50PN-derived blastocyst group were significantly lower than those in Day 60PN-derived blastocysts group(P<0.05).There were no significant differences in infertility years,BMI,basal hormone(FSH,LH,E_(2))levels,sperm normal morphology rate and acrosin activity between the two groups(P>0.05).The high-quality rate of Day 50PN-derived blastocysts group was significantly higher than that of Day 60PN-derived blastocysts group(P<0.01).There were no significant differences in the euploidy rate and the advanced age rate of women(≥40 years old)between the two groups(P>0.05).At the same ploidy,age and blastocyst quality,the mtDNA level of Day 50PN-derived blastocysts group was higher compared to Day 60PN-derived blastocysts group,but the differences were not significant(P>0.05).Conclusions:The mtDNA level could be a key factor that affects the developmental speed of 0PN-derived blastocysts.Lower mtDNA level is associated with delayed development of 0PN-derived blastocyst.However,more research is needed to verify it.
