摘  要:以南荻为原料,研究甲酸(formic acid,FA)联合碱性过氧化氢(alkaline hydrogen peroxide,AHP)处理对南荻纤维素酶解特性的影响,并采用傅里叶红外光谱、X射线衍射光谱和扫描电镜对处理前后的样品进行理化结构表征。研究结果表明,甲酸联合碱性过氧化氢处理获得的南荻AHP纤维素中纤维素含量显著提高至89.85%,且木质素和半纤维素含量大幅降低,分别为3.49%和3.72%。与南荻FA纤维素相比,南荻AHP纤维素甲酰化修饰消失、结晶度指数降低,更多的纤维素被暴露出来,使其纤维素酶解转化效率大幅提高。5%底物浓度时,酶解120 h南荻AHP纤维素酶解转化率可达95.54%,底物浓度为20%时,酶解液中葡萄糖浓度可达133.17 g/L,明显高于南荻原料和南荻FA纤维素。该研究结果为南荻纤维素高效转化制备清洁能源提供了技术支持和理论依据。Miscanthus lutarioriparius was used as raw material to study the effects of formic acid(FA)combined with alkaline hydrogen peroxide(AHP)on the enzymatic hydrolysis characteristics of cellulose from M.lutarioriparius,and the physicochemical structures of the samples before and after treatment were characterized by Fourier transform infrared spectroscopy,X-ray diffraction spectroscopy,and scanning electron microscopy.The results showed that the cellulose content of M.lutarioriparius AHP cellulose obtained by FA combined with AHP was significantly increased to 89.85%,whereas the content of lignin and hemicellulose was significantly decreased to 3.49%and 3.72%,respectively.As compared with M.lutarioriparius FA cellulose,the formylation modification disappeared,the crystallinity index decreased,and more cellulose was exposed in M.lutarioriparius AHP cellulose,which greatly improved the transformation efficiency of enzymatic hydrolysis.The conversion rate of M.lutarioriparius AHP cellulose reached 95.54%after 120 h at the substrate concentration of 5%.When the substrate concentration was 20%,the glucose concentration in the enzymatic solution was up to 133.17 g/L,which was significantly higher than that of raw material and M.lutarioriparius FA cellulose.The results of this study provide technical support and a theoretical basis for the efficient conversion of M.lutarioriparius cellulose to clean energy.
