摘 要:目的:建立紫外-可见分光光度法测定灵芝菌丝体中三萜及甾醇含量的方法。方法:以齐墩果酸为对照品,5%香草醛-冰醋酸和高氯酸为显色剂,以546nm波长测定灵芝菌丝体中三萜及甾醇含量。结果:灵芝菌丝体中三萜及甾醇质量在0~0.169 12mg范围内线性关系良好(R^(2)=0.999 3),精密度RSD为0.73%。样品溶液在配制后1h内稳定性较好,平均加标回收率为101.99%,RSD为1.44%。结论:该方法方便快捷,重现性好,结果可靠,可用于测定灵芝菌丝体中三萜及甾醇含量。Objective:To investigate the applicability of UV-vis spectrophotometry for the determination of triterpenoids and sterols in Ganoderma lucidum mycelium.Methods:The contents of triterpenes and sterols in Ganoderma lucidum mycelia were determined at 546nm with oleanolic acid as reference substance,5% vanillin aldehyde-glacial acetic acid and perchloric acid as chromogenic agent.Results:The triterpenoid and sterol in Ganoderma lucidumshowed a good relationship with in the range of 0~0.169 12mg(R^(2)=0.999 3),precision RSD was 0.73%.The sample solution has good stability within 1hour after preparation,the average recovery rate was 101.99%,and RSD was 1.44%.Conclusion:The method was convenient and fast.It has good reproducibility and reliable results.It can be used to determine the content of triterpenoids and sterols in Ganoderma lucidum mycelium.
