摘 要:目的观察土茯苓提取物对高尿酸致痛风大鼠的治疗效果。方法选取35只6周龄SPF级断乳SD雄性大鼠,随机分为空白对照组、模型组及土茯苓低、中、高剂量组,每组7只。除空白对照组外,其他4组大鼠均予以氧嗪酸钾灌胃4周,建立高尿酸致痛风大鼠模型,建模成功后空白对照组、模型组大鼠予以纯净水灌胃,持续4周;土茯苓低、中、高剂量组大鼠每日分别予以土茯苓1 mg/kg、10 mg/kg、100 mg/kg灌胃,持续4周。比较5组大鼠治疗前及治疗4周后血常规[包括白细胞计数(WBC)、嗜碱性细胞、中性粒细胞、嗜酸性细胞、淋巴细胞、单核细胞、红细胞计数(RBC)、血红蛋白(HGB)]、实质性脏器系数比值及血脂指标[总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)]。结果与空白对照组相比,模型组WBC、中性粒细胞、嗜酸粒细胞、淋巴细胞、单核细胞、肝脏指数及TC、TG、LDL-C升高(P<0.05);与模型组比较,土茯苓低、中、高剂量组中性粒细胞、嗜酸粒细胞、肝脏指数及TC、TG均降低,土茯苓低、高剂量组单核细胞均降低,土茯苓高剂量组LDL-C降低(P<0.05)。结论土茯苓对高尿酸致痛风具有明显的抗炎、降脂作用,且可减轻肝损伤。Objective To study the effect of smilax glabra for gout rats induced by hyperuricemia.Methods Thirty-five 6-week-old SPF weaned SD male rats were selected,and they were randomly divided into blank control group,model group and low,middle and high-dose smilax glabra groups,7 animals in each group.Except the blank control group,the other four groups of rats were given potassium oxazinate by gavage for 4 weeks to establish gout rat model induced by hyperuricemia.After successful modeling,rats in the blank control group and model group were given purified water by gavage for 4 weeks.Rats in the low,middle and high-dose smilax glabra groups were given 1 mg/kg,10 mg/kg and 100 mg/kg of smilax glabra by gavage every day for 4 weeks.Blood count(WBC,basophils,neutrophils,eosinophils,lymphocytes,monocytes,RBC,HGB),ratio of parenchymal organ coefficient and blood lipid indexes(TC,TG,LDL-C,HDL-C)before and after 4 weeks of treatment were compared among 5 groups.Results Compared with the blank control group,WBC,neutrophils,eosinophils,lymphocytes,monocytes,liver index,TC,TG and LDL-C in the model group increased(P<0.05).Compared with the model group,neutrophils,eosinophils,liver index,TC and TG in low,middle and high-dose smilax glabra group decreased,monocytes in low,high-dose smilax glabra groups decreased,and LDL-C in high-dose smilax glabra group decreased(P<0.05).Conclusion Smilax glabra has obvious anti-inflammatory and lipid-lowering effects on gout induced by hyperuricemia,and can alleviate liver injury.
