摘 要:禽腺病毒(Fowl Adenovirus,FAdV)是一组血清型众多及致病性多样的病原体,多数呈亚临诊性长期潜伏带毒,部分毒株可造成禽群包涵体肝炎、心包积液、肌胃糜烂症、产蛋下降、生长迟缓等病症,严重威胁养禽业健康发展。为分析血清10型禽腺病毒(FAdV-10)全基因组序列特征,本研究将分离自外表健康鸡泄殖腔拭子的FAdV-10 AHFY19株经卵黄囊途径接种8日龄SPF鸡胚扩繁,从死亡鸡胚肝组织中提取病毒基因组,采用illumina二代基因测序技术和特殊结构区二温式PCR扩增测定病毒全基因组并分析其遗传特征。结果显示,AHFY19株(103.1EID50/0.1 mL/枚)接种的鸡胚于18~21日龄全部死亡。全基因组测序显示,AHFY19株病毒基因组全长为45757 bp(GenBank登录号:MN542422),其G+C含量为54.63%,末端重复序列(ITR)长为56 bp,共有52个开放阅读框(ORF)。基因组比对分析显示,AHFY19株属I群FAdV C种FAdV-10型,与C2-B-FAdV-10株(MK572851)全基因组同源性最高(99.91%),但AHFY19株基因组多203 bp,差异位于fiber-1、fiber-2和ORF19A基因,然而这些差异并不存在于FAdV-10型C-2B株。AHFY19株与FAdV-4毒株全基因组核苷酸序列同源性在98.35%~98.58%之间。主要结构蛋白基因遗传进化分析显示,仅hexon基因发育进化树可区分C种FAdV-10型与FAdV-4型毒株。氨基酸分析显示,AHFY19株Hexon蛋白V188、Fiber-2蛋白G219和A380与FAdV-4型弱毒株一致。本研究为国内首次测定公布FAdV-10型病毒全基因组序列,其基因组分子特征为深入分析FAdV遗传演化提供了研究资料。Fowl adenoviruses(FAdVs)are pathogens with numerous serotypes and diverse types of pathogenicity.They are a seriously threat to development of the poultry industry worldwide.FAdV infection is characterized by hepatitis,pericardial effusion,gizzard erosion,reduced egg production,and growth retardation.We wished to characterize the whole genome sequence of FAdV-10.The AHFY19 strain isolated from the cloacal swabs of externally healthy chickens was inoculated into 8-day-old specific pathogen-free chicken embryos through yolk sacs for viral propagation.The viral genome was extracted from the liver tissues of dead chicken embryos.The whole genome of the virus was sequenced and analyzed by next-generation genesequencing technology(illumina)and amplification by two-temperature polymerase chain reactions of specific structural regions.We discovered that all chicken embryos inoculated with the AHFY19 strain(103.1EID50/0.1mL/piece)died at 18–21 days of age.Whole-genome sequencing showed that the full-length genome of the AHFY19 strain was 45757 bp(GenBank accession number:MN542422)with 54.63%G+C content,56 bp of inverted terminal repeats,and 52 open reading frames.Genome comparison showed that the AHFY19 strain belonged to the C species of group-I FAdV-10 and had the highest genome homology(99.91%)with the C2-BFAdV-10 strain(MK572851).The genome of the AHFY19 strain was 203-bp more than that of the C2-B-FAdV-10 strain in fiber-1,fiber-2,and ORF19A.However,these differences were not present in the FAdV-10 type C-2B strain.The homology of genome nucleotide sequences between AHFY19 and FAdV-4 strains was 98.35%–98.58%.Genetic-evolution analysis of the genes of major structural proteins showed that C species FAdV-10 and FAdV-4 strains could be distinguished only by the phylogenetic tree of hexon.Amino-acid analysis showed that the Hexon protein V188,Fiber-2 protein G219,and A380 of the AHFY19 strain were consistent with the FAdV-4-attenuated strain.In conclusion,we published,for the first time,the whole genome sequence of
