以乙醇检测为例分析溶液温度对空气置换式移液器移液量的影响

摘  要:以乙醇检测为例,参照《GA/T 842-2019血液酒精含量的检验方法》的检测方法,考察了4℃、24℃两个不同温度的内标工作液(叔丁醇)用于乙醇含量检测时结果的差异,发现了溶液温度会影响空气置换式移液器的移液量。分别使用不同润枪方式移取不同温度的溶液并称量移取量,找到了三个引起上述差异的因素:润枪环节、溶液密度、溶液粘度。经统计分析,上述三个因素的贡献程度由高到低依次是:润枪环节中低温内标工作液对移液器吸头和腔体内空气密度的改变从而导致移取液体时产生的吸力减小、低温内标工作液本身的密度、低温内标工作液本身的粘度。The difference was investigated in the results of using internal standard working solutions(t-butanol)at 4℃and 24℃for the determination of alcohol content with reference to GA/T 842-2019 Examination Methods for Ethanol Concentration in blood of Industry Standard of Public Security of China.It was found that the ethanol detection value with the internal standard at 4℃was higher.Pipetting solution at different temperatures by different ways of rinsing,3 influencing factors were found as follows rinsing,solution density and solution viscosity.According to statistical analysis,the contribution of the above three factors from high to low was as follows the low-temperature solution rinsing link to the change of the density of the air in the pipette suction head and cavity which led to the decrease of the suction force when the liquid was pipetted,the density of the low-temperature solution,and the viscosity of the low-temperature solution.
