摘 要:【目的】探究GLK基因家族在黄叶型连翘叶片呈色及响应光照度中的作用。【方法】通过生物信息学方法鉴定连翘GLK家族成员,分析其系统进化、基因结构和启动子顺式作用元件,结合转录组数据和实时荧光定量PCR技术筛选与连翘叶色相关及响应光照度的关键基因。【结果】从连翘基因组中鉴定到44个GLK基因家族成员,命名为FsGLK1~FsGLK44,分为5个亚族,同一亚族具有相似的保守结构域,所有FsGLK基因的启动子区域均具有多个光响应元件。转录组数据表明,5个FsGLK基因在绿叶和黄叶型连翘中差异表达,其中FsGLK9、FsGLK19、FsGLK27在绿叶型连翘中优势表达,FsGLK34、FsGLK41在黄叶型连翘中优势表达。FsGLK19、FsGLK32、FsGLK41在黄叶型连翘全光和遮光处理条件下差异表达。结合实时荧光定量分析发现,FsGLK19明显受黑暗条件诱导表达。【结论】连翘中有44个FsGLK基因,其中FsGLK19可能是黄叶型连翘呈色及响应光照度的关键基因。【Objective】The purpose of this study is to explore the role of GLK gene family in the color presentation and response to light intensity of yellow-leafed Forsythia.【Methods】The GLK family members of Forsythia suspensa were identified by bioinformatics.The phylogeny,gene structure and cis acting elements of promoter were analyzed.Combined with transcriptome data and qRT-PCR,the key genes related to leaf color and response to light intensity of Forsythia were screened.【Results】A total of 44 GLK family members were identified from Forsythia suspensa’s genome,which were named FsGLK1-FsGLK44.These 44 GLK family members were divided into five subgroups,each of which had similar conserved domains.The promoter region of all FsGLK genes had multiple light-responsive elements.Transcriptome data showed that 5 FsGLK genes had different expression in green-leafed and yellow-leafed Forsythia.Among them,FsGLK9,FsGLK19 and FsGLK27 were preferentially expressed in green-leafed Forsythia.While,FsGLK34 and FsGLK41 were preferentially expressed in yellow-leafed Forsythia.FsGLK19,FsGLK32 and FsGLK41 had different expression under full light and shading treatments in yellow-leafed Forsythia.Combined with qRT-PCR analysis,FsGLK19 was found to be significantly induced by dark conditions.【Conclusion】There were 44 FsGLKs in Forsythia suspensa,among which FsGLK19 may be a key gene for the color presentation and light intensity response in yellow-leafed Forsythia.
