摘 要:【目的】为准确、快速地鉴别桃铃型花和蔷薇型花,开发与桃花型性状相关的dCAPs标记。【方法】根据花型性状基因定位结果,其关联度最高的SNP位点位于PpB3-1基因启动子区域,基于SNP位点开发含有内切酶SalI-HF的dCAPs标记,在1个桃杂交群体(73个杂种单株)和1个桃自然群体(102份种质资源)中扩增,用SalI-HF限制性内切酶对特异扩增片段进行酶切,用聚丙烯酰胺凝胶电泳的方式对酶切产物进行检测分析。【结果】铃型花DNA扩增出来的条带可以被酶切,产生两种类型特异片段:308和282 bp两条特异片段(杂合显性,A/C)和282 bp一条特异片段(纯合显性,C/C);蔷薇型花DNA扩增出来的条带不能被酶切,产生一条308 bp特异片段(纯合隐性,A/A)。该标记在桃杂交群体和自然群体中的检测符合率均为100%。【结论】桃花型性状的dCAPs标记B3-1可用于桃花型性状分子标记辅助选择育种。【Object】In order to accurately and quickly identify showy(sh)and non-showy(SH)flower of peach,dCAPs markers related to flower-type trait in peach were developed in this study.【Methods】According to the gene mapping results of flower-type trait,the SNP that highly associated of flower types was located inthe promoter region of PpB3-1.Based on the SNP sites,the dCAPs marker(B3-1)containing endonuclease SalI-HF was developed and amplified in one peach F1 population(73 hybirds)and one peach natural populations(102 germplasm resources).The specific amplified fragments were digested by enzyme SalI-HF,and the digested products were detected by polyacrylamide gel electrophoresis.【Results】The amplified band of non-showy flower DNA can be digested to produce two types of specific fragments:two specific fragments of 308 bp and 282 bp(heterozygous dominant,A/C)and one specific fragment of 282 bp(homozygous dominant,C/C);the amplified band of showy flower DNA cannot be digested,resulting in producing a 308 bp specific fragment(homozygous recessive,A/A).The detection coincidence rate of the dCAPs marker reached 100%both in 73 peach hybrids and 102 peach accessions.【Conclusion】The dCAPs marker B3-1 can be used for molecular marker-assisted breeding of theidentification of peach flower type.
