摘  要:目的探讨大鼠海马组织星形胶质细胞活化水平及炎症因子表达在急性一氧化碳中毒迟发性脑病(DEACMP)中的作用及可能的机制。方法经Morris水迷宫训练筛选认知功能正常的SD雄性大鼠,随机分为实验组,对照组,实验组大鼠采用静态吸入法置备DEACMP模型。分别于中毒后7d、14d、21d各时间点通过Morris水迷宫比较两组学习记忆能力;苏木精-伊红染色观察海马病理损伤程度;免疫荧光法和Western blot法检测海马区星形胶质细胞特异性标志物(GFAP)的表达情况;ELSIA法检测海马组织中TNF-α、IL-1β、IL-6表达水平。结果①大鼠表现出典型的CO中毒症状;②Morris水迷宫结果表明,实验组第7天大鼠水迷宫逃避潜伏期无明显差异(P>0.05);实验组14d、21d水迷宫逃避潜伏期逐渐延长,与对照组相比差异有统计学意义(t=2.60、t=5.56,P<0.05);③苏木精-伊红染色与对照组相比,实验组海马细胞出现不同程度的细胞损伤;④免疫荧光结果统计显示,实验组第7天、14天、21天的海马CA1区GFAP表达明显增多,与对照组相比差异有统计学意义(t=-5.20、t=-5.38,t=-6.57,P<0.05);⑤Western blot结果显示,实验组海马组织中GFAP蛋白表达显著升高,在7d(1.37±0.07)、14d(1.35±0.16)、21d(1.35±0.14)较对照组(1.00±0.00)差异有统计学意义(t=-8.81、t=-3.80、t=-4.34、P<0.05);⑥ELISA结果表明,实验组各组TNF-α、IL-1β、IL-6蛋白表达明显升高,分别在在7d(t=-6.23、t=-5.99、t=-5.30)、14d(t=-4.68、t=-3.86、t=-4.96)、21d(t=-4.97、t=-7.31、t=-3.44)较对照组有统计学意义(P<0.05)。结论大鼠海马区星形胶质细胞活化水平及炎症因子TNF-α、IL-1β、IL-6表达水平与DEACMP的病理发展具有相关性。Objective To explore the role and possible mechanism of activation level of astrocytes and expression of inflammatory cytokines in hippocampus of rats in delayed encephalopathy after acute carbon monoxide poisoning(DEACMP).Methods After the Morris water maze training,SD male rats with normal cognitive function were screened and randomly divided into the experimental group and the control group.The rats in the experimental group were prepared with the DEACMP model by static inhalation.The learning and memory abilities of the two groups were compared by Morris water maze at each time point 7 d,14 d,and 21 d after poisoning;Hematoxylin-eosin staining was used to observe the degree of pathological damage to the hippocampus;Immunofluorescence and Western blot were used to detect the expression of astrocyte specific marker(GFAP)in hippocampus.The expression levels of TNF-α,IL-1βand IL-6 in hippocampus were detected by ELISA.Results①The rats showed typical symptoms of CO poisoning;②Morris water maze results showed that there was no significant difference in the escape latency of the experimental group on the 7 d(P>0.05),The escape latency of water maze in experimental group was gradually prolonged on 14 d and 21 d,and the difference was statistically significant compared with that in control group(t=2.60,t=5.56,P<0.05);③Compared with the control group,Hematoxylin-eosin staining showed different degrees of cell damage in hippocampus of experimental group;④The results of immunofluorescence showed that the expression of GFAP in hippocampal CA1 area of experimental group increased significantly on 7 d,14 d and 21 d,and the differences were statistically significant compared with those of control group(t=-5.20,t=-5.38,t=-6.57,P<0.05);⑤The results of Western blot showed that the expression of GFAP protein in hippocampus of experimental group was significantly increased,and there was a statistical difference in 7 d(1.37±0.07),14 d(1.35±0.16)and 21 d(1.35±0.14)compared with that of control group(1.00±0.00),(t=-8
