摘 要:目的:探讨跑台运动干预对帕金森病(PD)模型小鼠黑质及其投射的海马CA1区、海马下托神经元计数及多巴胺转运体(DAT)、脑源性神经营养因子(BDNF)和酪氨酸激酶受体B(TrkB)表达的影响。方法:24只8周龄雌性C57BL/6小鼠随机分为生理盐水对照组(SC组)、PD模型组(PD组)、生理盐水+有氧运动组(SE组)、PD+有氧运动组(PDE组),每组6只,其中,PD和PDE组小鼠按照30 mg/kg剂量每隔3.5天腹腔注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)溶液一次,SC和SE组注射同等剂量生理盐水,共5周。SE组和PDE组进行7周跑台运动干预,运动方案为:12 m/min,60 min/天,5天/周。干预结束后48小时取材并采用尼氏染色法观察黑质、海马CA1区和海马下托神经元计数等,采用免疫荧光技术观察DAT、BDNF和TrkB在黑质、海马CA1区和海马下托中的表达情况。结果:(1)双因素方差分析表明,跑台运动和MPTP注射对小鼠黑质神经元计数、DAT、BDNF和TrkB表达,海马CA1区神经元计数、DAT、TrkB表达,以及海马下托神经元计数、DAT、TrkB表达均无交互作用(P>0.05);对海马CA1区和海马下托BDNF表达具有交互作用(P<0.05)。(2)与SC组相比,PD组小鼠黑质神经元数量显著减少;与PD组相比,PDE组小鼠黑质神经元数量显著增加(P<0.01)。(3)与SC组相比,PD组黑质和海马下托DAT蛋白表达均显著降低;PDE组比PD组小鼠黑质DAT表达显著升高(P<0.01);(4)与PD组相比,PDE组小鼠黑质和海马CA1区BDNF均显著升高(P<0.01),海马下托BDNF和TrkB表达存在上升的趋势,但不显著。结论:跑台运动可以改善MPTP染毒导致的小鼠黑质海马CA1区和海马下托中DAT、BDNF和TrkB蛋白表达下调,提示可能是运动改善PD症状的原因之一。Objective To explore the effect of treadmill exercise on the expression of dopamine transporter(DAT),brain-derived neurotrophic factor(BDNF)and tyrosine kinase receptor B(TrkB)in substantia nigra,CA1 area of hippocampus and subiculum of mice with Parkinson’s disease(PD).Methods Twenty-four 8-week-old female C57BL/6 mice were randomly divided into a normal saline control group(SC group),a PD model group(PD group),a normal saline+aerobic exercise group(SE group)and a PD+aerobic exercise group(PDE group),each of six.The mice of PD and PDE groups were intraperitoneally injected with MPTP solution at a dose of 30 mg/kg every 3.5 days,while those of SC and SE groups were injected with the normal saline in the same way for 5 weeks.Moreover,the SE and PDE groups underwent 60-minute treadmill running at 12 m/min 5 days a week for 7weeks.Then,48 hours after the intervention,Nissl staining was used to observe the number of neurons in substantia nigra,CA1 region of hippocampus and subiculum,while immunofluorescence was employed to observe the expression of DAT,BDNF and TrkB in the same place.Results(1)Two-way ANOVA showed that treadmill exercise and MPTP injection had no interaction effect on the number of neurons and the expression of DAT and TrkB in nigra,hippocampal CA1 region and subiculum(P>0.05),but had an interaction effect on the expression of BDNF in CA1 region of hippocampus and subiculum(P<0.05).(2)The number of substantia nigra neurons in PD group decreased significantly compared with group SC and PD group(P<0.01).(3)Compared with SC group,the protein expression of DAT in substantia nigra and subiculum of PD group decreased significantly,while the expression of DAT in the substantia nigra of PDE group was significantly higher than that of PD group(P<0.01).(4)Compared with PD group,a significant increase was observed in BDNF in substantia nigra and CA1region of hippocampus in PDE group(P<0.01),while the expressions of BDNF and TrkB in the subiculum showed an upward trend,but not significant.Conclusion Treadmill
