摘 要:目的 通过构建慢性哮喘大鼠模型,研究白头翁皂苷B4对慢性哮喘大鼠肺组织中转化生长因子-β1(transforming growth factor β1,TGF-β1)、p38丝裂原活化蛋白激酶(mitogen activated protein kinase,p38 MAPK)的调控作用和氧化应激水平的影响。方法 构建慢性哮喘大鼠模型,将72只SD大鼠随机分为健康对照组、慢性哮喘组、地塞米松组、白头翁皂苷B4高剂量组、白头翁皂苷B4低剂量组、白头翁皂苷B4+TGF-β1激活组,每组12只。检测各组大鼠肺泡灌洗液中各种类细胞数量;苏木精-伊红(hematoxylin-eosin,HE)染色法检测各组大鼠肺组织病理学变化;试剂盒检测各组大鼠肺组织中超氧化物歧化酶(superoxide dismutases,SOD)、丙二醛(malondialdehyde,MDA)含量及炎症因子白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素6(interleukin-6,IL-6)、血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平;Western blot法检测TGF-β1、p38MAPK、p-p38MAPK蛋白的表达。结果 健康对照组大鼠肺组织结构完整;与健康对照组相比,慢性哮喘组大鼠出现明显肺损伤,肺泡灌洗液中细胞总数、中性粒细胞、嗜酸性粒细胞、淋巴细胞数量升高,肺组织MDA[(0.96±0.15)vs(19.36±1.79)μmol/L]、IL-1β、IL-6、TNF-α含量和TGF-β1蛋白表达量[0.21±0.06 vs 2.35±0.12]、p-p38MAPK/p38MAPK[0.25±0.07 vs 0.88±0.06]升高,SOD含量[(159.51±6.37)vs(42.15±5.73)U/L]降低,差异均有统计学意义(均P<0.05);与慢性哮喘组相比,地塞米松组、白头翁皂苷B4高剂量组、白头翁皂苷B4低剂量组大鼠肺组织逐渐恢复,肺泡灌洗液中细胞总数、中性粒细胞、嗜酸性粒细胞、淋巴细胞数量降低,肺组织MDA[(19.36±1.79)vs(3.48±0.16)、(3.53±0.31)、(14.25±0.65)μmol/L]、IL-1β、IL-6、TNF-α含量和TGF-β1蛋白表达量[(2.35±0.12)vs(0.57±0.08)、(0.61±0.11)、(1.15±0.12)]、p-p38MAPK/p38MAPK[0.88±0.06 vs 0.41±0.07、0.39±0.05、0.62±0.09]降低,SOD含量[(42.15±5.73)vs(131.62Objective By constructing a rat model of chronic asthma,to explore the regulatory effect of Pulsatilla saponin B4 on transforming growth factor-β1(TGF-β1) and p38 mitogen-activated protein kinase(p38MAPK) in the lung tissue of chronic asthmatic rats and its influence on oxidative stress levels.Methods A rat model of chronic asthma was constructed,and 72 SD rats were randomly separated into healthy control group,chronic asthma group,dexamethasone group,high-dose Pulsatilla saponin group,low-dose Pulsatilla saponin group,Pulsatilla saponin +TGF-β1 activated group,12 rats per group.The number of various types of cells in the bronchoalveolar lavage fluid of rats in each group was measured.The hematoxylin-eosin(HE) was performed to measure the pathological changes of lung tissue of rats in each group.The kits were applied to measure the levels of superoxide dismutase(SOD),malondialdehyde(MDA) and related inflammatory factors,including interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α),in lung tissue of rats in each group.Western blot was performed to measure the expression of TGF-β1,p38MAPK and p-p38MAPK proteins.Results The lung tissue structure of the rats in the healthy control group was intact.Compared with the healthy control group,the rats in the chronic asthma group developed significant lung damage,the total number of cells,and the number of neutrophils,eosinophils and lymphocytes in alveolar lavage fluid increased,the content of MDA [(0.96 ±0.15) vs(19.36 ±1.79)μmol/L],the levels of IL-1β,IL-6 and TNF-α,the expression of TGF-β1 [(0.21 ±0.06) vs(2.35±0.12)],and p-p38MAPK/p38MAPK[0.25±0.07 vs 0.88±0.06] in lung tissue increased,the content of SOD[(159.51±6.37) vs(42.15 ±5.73)U/L] decreased,and the differences were statistically significant(all P <0.05).Compared with the chronic asthma group,the lung tissue of the rats in the dexamethasone group,the high-dose Pulsatilla saponin group,and the low-dose Pulsatilla saponin group gradually recovered,the total number of cel
