摘 要:为了进一步提高甜椒核雄性不育两用系育种效率,利用AB91核雄性不育候选基因Capana05g000747的突变位点设计KASP分子标记,以转育甜椒核雄性不育两用系的F_(2)群体及育成的两用系为试验材料,对植株的育性进行基因型检测,并检验该标记的准确性。结果表明,该分子标记与甜椒核雄性不育的育性呈现共分离,在苗期对F_(2)植株的基因型进行早期鉴别,分型准确率分别为98.91%和99.04%,可有效淘汰基因型为MSMS的可育株,筛选出基因型为MSms的可育株直接用于不育系的转育,转育出了黄色和红色甜椒核雄性不育两用系AB91-HTJ412和AB91-HLD163。在苗期对两用系群体的育性基因型进行早期鉴别,可有效淘汰基因型为MSms的可育株,选留基因型为msms的不育株直接用于杂交组合选配及杂交制种。In order to further improve the breeding efficiency of genic male sterile line dual purpose system of sweet pepper,KASP molecular markers were designed by using the mutation site of the‘AB91’genic male sterile candidate gene Capana05g000747.The F_(2) population and dual purpose system of transformation sweet pepper genic male sterile lines were used as test materials to detect the genotype of plant fertility and check the accuracy of this marker.The results indicated that the molecular marker presented common separation with the fertility of genic male sterile sweet pepper.Early identification was carried out on genotype of F_(2) plant.The classification accuracy were 98.91%and 99.04%,respectively.The effective eliminated genotype was the fertile plant of MSMS,which could be directly used for transformation of sterile line.The dual purpose system of yellow and red genic male sterile lines sweet peper‘AB91-HTJ412’and‘AB91-HLD163’were turn out.During seedling period,early stage identification on fertility genotypes of dual purpose system population could effectively eliminate the fertile plants of MSms genotype.And the selected and remaining genotype was sterile plants of msms,which could be directly used in cross combination breeding and hybrid seed production.
