摘 要:目的比较小鼠阴道和外周血γδT细胞表型特征和细胞因子等异同,为阴道γδT细胞的功能研究奠定基础。方法分离小鼠阴道和外周血单个核细胞,流式细胞术检测γδT细胞比例、趋化因子受体、记忆分子、杀伤分子和ERα的表达差异。PMA+Ionomycin刺激后检测细胞因子IL-17和IFN-γ的表达,并分析不同组织γδT细胞产生的IL-17量与ERα密度的关系。结果阴道内富含γδT细胞,γδT细胞占阴道T比例为47.91%±3.12%,主要是组织定居记忆γδT细胞亚型,高表达CD44、CD69、CD103、IL-17,但低表达IFN-γ。此外,脾、外周血、肺、膀胱、子宫、阴道的γδT细胞IL-17表达量与其ERα密度呈正相关。结论阴道γδT细胞主要为组织定居记忆γδT细胞,阴道γδT细胞可能通过产生大量IL-17在抵抗生殖道病原微生物感染中发挥重要功能。This study was performed to compare the phenotype and cytokines ofγδT cells from mouse vagina and blood,for lay a foundation for further research of the immune function of vaginalγδT cells.The mononuclear cells from vagina and blood were isolated.Flow cytometry was used to detect the percentage ofγδT cells,the expression of chemokine receptors,memory markers,cytotoxicity molecules and Erα;the expression of IL-17 and IFN-γwere detected after PMA+Ionomycin stimulation;the correlation between the amount of IL-17 produced byγδT cells and ERαdensity was analyzed.Data showed that the percentage ofγδT cells in vaginal T cells was47.91%±3.12%,andγδT cells in vagina were mainly tissue resident memoryγδT cells with high expression of CD44,CD69,CD103 and IL-17,but low expression of IFN-γ.In addition,the expression of IL-17 in spleen,peripheral blood,lung,bladder,uterus and vagina were positively correlated with ERαdensity.In conclusion,vaginalγδT cells were mainly tissue resident memoryγδT cells which might play an important role in bacterial infection by producing IL-17.
