健脾行气活血方配合中药香包治疗糖尿病胃轻瘫效果研究

摘　　要：目的探讨健脾行气活血方(自拟糖胃方)配合中药香包在糖尿病胃轻瘫治疗中的效果。方法购置健康SD大鼠20只,雌雄不限,4~6周鼠龄,常规分离、培养平滑肌细胞,常规制备细胞悬液,完成细胞密度调整后均接种在6孔板中,并将细胞随机分为正常对照组、高糖组、中药组与JNK抑制剂组,各组均给予相应的措施干预3 d,比较各组细胞形态、平滑肌细胞收缩、细胞自噬溶酶体形成及自噬相关分子。结果中药组、JNK抑制剂组细胞形态基本规则,且中药组细胞形态略优于JNK抑制剂组;中药组平滑肌细胞平均长度高于JNK抑制剂组与高糖组,差异有统计学意义(P<0.05);收缩率低于JNK抑制剂组与高糖组,差异有统计学意义(P<0.05);JNK抑制剂组与中药组可见2个自噬体,但中药组线粒体肿胀、空泡化及内质网扩张轻于JNK抑制剂组,差异有统计学意义(P<0.05);中药组LC3-Ⅱ、Beclin-1及JNK低于JNK抑制剂组,差异有统计学意义(P<0.05)。结论健脾行气活血方(自拟糖胃方)配合中药香包用于糖尿病胃轻瘫治疗中可作用于平滑肌细胞,能通过调节自噬相关分子水平,发挥平滑肌细胞自噬作用。Objective To explore the effect of Jianpi Xingqi Huoxue Recipe(self-made Tangwei Recipe) combined with traditional Chinese medicine sachets in the treatment of diabetic gastroparesis.Methods Twenty healthy SD rats(male or female,4-6 weeks old)were used to routinely isolate and culture smooth muscle cells,routinely prepare cell suspensions,and inoculate them in 6-well plates after adjusting the cell density.These cells were randomly divided into normal control group,high glucose group,traditional Chinese medicine group and JNK in-hibitor group.Each group was given corresponding measures to intervene for 3 days,and the cell morphology,smooth muscle cell contraction,autophagy-lysosome formation and autophagy-related molecules in each group were compared.Results The morphology of cells in the traditional Chinese medicine group and the JNK inhibitor group was basically regular,and the cell morphology in the traditional Chinese medicine group was slightly better than that in the JNK inhibitor group.The average length of smooth muscle cells in the traditional Chinese medicine group was higher than that in the JNK inhibitor group and the high glucose group,and the difference was statistically signifi-cant(P<0.05).The contraction rate in the traditional Chinese medicine group was lower than that of the JNK in-hibitor group and the high glucose group,and the difference was statistically significant(P<0.05).The vesicles and endoplasmic reticulum expansion were lighter than those in the JNK inhibitor group,and the difference was statisti-cally significant(P<0.05).Conclusion Jianpi Xingqi Huoxue Recipe(self-made Tangwei Recipe)combined with traditional Chinese medicine sachets can act on smooth muscle cells in the treatment of diabetic gastroparesis and can play a role in smooth muscle cell autophagy by regulating the level of autophagy-related molecules.
