摘 要:目的研究医院制剂复方草豆蔻合剂质量标准中干姜薄层色谱鉴别方法的耐用性。方法采用薄层色谱法鉴别复方草豆蔻合剂中的干姜,并考察不同厂家薄层板,展开剂预饱和与否,不同湿度,不同温度条件下薄层鉴别方法的耐用性。结果Merck高效硅胶预制板和硅胶预制板图谱斑点清晰,分离效果好;硅胶G自制硅胶板图谱主斑点清晰,分离效果好。展开剂预饱和15 min的图谱和未饱和的图谱斑点都很清晰,分离效果都很好。在双槽展开缸的一侧加不同浓度的硫酸,湿度18%(硫酸∶水=100∶95.5)薄层色谱与湿度88%(硫酸∶水=10.8∶100)相比较为清晰,分离效果相近。低温下薄层色谱,色谱斑点分离效果不好,对主斑点有影响。结论所建立的干姜薄层色谱鉴别方法简便、专属性强、耐用性好,可作为复方草豆蔻合剂的质量控制方法。Objective To study the durability of the thin layer chromatographic identification of rhizoma zingiberis in the quality standard of compound alpinia katsumadai mixture.Methods The rhizoma zingiberis in compound alpinia katsumadai mixture was identified by thin layer chromatography,and the durability of the thin layer identification method was investigated under the conditions of different manufacturers'thin layer plates,whether the developing agent was pre saturated or not,different humidity and different temperature.Results Merck high-efficiency silica gel prefabricated plate and silica gel prefabricated plate had clear spots and good separation;silica gel G homemade silica gel plate had clear main spots and good separation.The chromatograms of the spreaders pre-saturated for 15 min and the unsaturated chromatograms had clear spots and good separation.The thin layer chromatogram at 18%humidity(sulfuric acid∶water=100∶95.5)was clearer and the separation was similar compared with 88%humidity(sulfuric acid∶water=10.8∶100)when different concentrations of sulfuric acid were added to one side of the double-slotted spreading cylinder.Thin layer chromatography under low temperature,chromatographic spots were not well separated and had an effect on the main spots.Thin layer chromatography under low temperature,chromatographic spots were not well separated and had an effect on the main spot.Conclusion The established thin-layer chromatographic identification of rhizoma zingiberis is simple,specific,and durable,and can be used as a quality control method for the compound alpinia katsumadai mixture.
