摘  要:以藤仓赤霉菌CICC 2444为出发菌株,采用常压室温等离子体(ARTP)诱变获得突变菌株,并根据赤霉素的代谢途径特征,选择真菌细胞壁合成抑制剂酮康唑作为筛选压力筛选赤霉素高产菌株。结果表明,经过抗性初筛和摇瓶复筛,获得一株赤霉素高产菌株3-6-1,GA_(3)产量达194.64 mg·L^(-1),较出发菌株提高了64%;经传代实验验证,该菌株遗传稳定性良好;经7.5 L发酵罐分批放大培养,该菌株的发酵终产量为235.98 mg·L^(-1),较出发菌株提高了87.2%,具备成为工业菌株的潜力。Using Fusarium fujikuroi CICC 2444 as an original strain,we obtained the mutant strain by atmospheric and room temperature plasma(ARTP)mutagenesis.Moreover,according to the characteristics of metabolic pathways of Gibberellin,we screened a high-yield Gibberellin-producing strain by using the fungal cell wall synthesis inhibitor ketoconazole as the screening pressure.The results show that a high-yield Gibberellin-producing strain 3-6-1 is obtained through preliminary screening and shake flask screening.The yield of GA_(3) reaches 194.64 mg·L^(-1),which is 64% higher than that of the original strain.The results of passage experiment show that the genetic stability of the strain 3-6-1 is good.The final fermentation yield of the strain 3-6-1 is 235.98 mg·L^(-1) after batch amplification in 7.5 L fermentor,which is 87.2% higher than that of the original strain,indicating the potential to become an industrial strain.
