摘  要:目的 基于UPLC-Q-TOF-MS建立椿乳凝胶化学成分快速分析方法。方法 采用Agilent C clipsePlus C_(18)(2.1 mm×100 mm,1.8 μm);流动相为0.1%甲酸水-乙腈,梯度洗脱;流速0.30 mL·min^(-1)。正负离子采集模式下进行全扫描质谱。通过MassBank、NIST等数据库和Qualitative Workflows B.08.00,并结合相关文献检索以及质谱裂解规律分析椿乳凝胶中的化学成分,使用GC-MS法分析制剂中的挥发性成分。结果 共鉴定出106个化合物,其中57个化合物来自椿皮(2个与苦参共有,2个与牡丹皮共有,2个与乳香共有),44个化合物来自苦参(2个与椿皮共有),4个化合物来自牡丹皮(2个与椿皮共有),5个化合物来自乳香(2个与牡丹皮共有),2个化合物来自冰片。结论 本研究快速、准确地阐明了椿乳凝胶的活性成分,为进一步研究椿乳凝胶及其临床合理应用提供参考。Objective To establish a method for the rapid analysis of chemical constituents in Chunru gel by UPLC-Q-TOF-MS.Methods Agilent C clipsePlus C_(18)(2.1 mm×100 mm,1.8 μm) was used to identify the chemical constituents in Chunru gel.The mobile phase was 0.1% formic acid water-acetonitrile with gradient elution at 0.30 mL·min^(-1).Full scan mass spectrometry was performed in positive and negative ion acquisition mode.With MassBank,NIST and other databases and Qualitative Workflows B.08.00 combined with relevant literature search and mass spectrometry fragmentation rules,the chemical components in Chunru gel were analyzed.GC-MS technology was used to analyze the volatile components in the preparation.Results Totally 106 compounds were identified by UPLC-Q-TOF-MS.Among them,57 compounds were from Ailanthus altissima Swingle (2 shared with Sophora flavescens,2 shared with Chinese moutan cortex,and 2 shared with Frankincense);44 compounds were from Sophora flavescens (2 shared with Ailanthus altissima Swingle);4 compounds were from Chinese moutan cortex (2 shared with Ailanthus altissima Swingle);5 compounds were from Frankincense (2 shared with Ailanthus altissima Swingle);and 2 compounds were from borneol.Conclusion This method can quickly and accurately elucidate the active components in Chunru gel,and provide a reference for its further research and its rational clinical application.
