摘 要:目的 采用高效液色谱相串联飞行时间质谱对姜半夏和姜虎掌进行化合物分析,用Mass Profiler Professional(MPP)统计软件查找特征化合物并建立数据模型。方法 采用Agilent InfinityLab Poroshell 120EC-C(4.6 mm×150 mm,3 μm)色谱柱,以甲醇-5 mmol乙酸铵梯度洗脱,流速0.4 mL·min^(-1);采用电喷雾离子化源,正离子扫描;Profinder软件以MFE算法寻找化合物,MPP软件根据姜半夏和姜虎掌各自化合物出现的频率、响应值、可变性、特征性等进行过滤分析,设定阈值为P<0.01,最小倍数变化值FC<2.0,寻找姜半夏和姜虎掌的差异化合物,并建立两者的模型。结果 初步鉴定到姜半夏特有成分12个,姜虎掌特有成分99个,建立了姜半夏和姜虎掌数据模型,在掺入一定比例姜虎掌时,模型能预测样品偏离,具有一定的预测性。结论 通过高分辨液质联用仪和MPP统计软件对半夏及其伪品虎掌的姜制炮制品的化学成分进行分析,找到了姜虎掌特有的成分,选择含量较高的作为姜半夏掺姜虎掌的鉴别成分,同时建立的数据模型具有一定的预测性,为半夏炮制品中掺伪检查提供一定的思路。Objective To analyse the components of ginger Pinellia ternata and ginger Pinellia pedatisecta Schott by Q-TOF-MS,and find the characteristic compounds and establish the data model by Mass Profiler Professional (MPP).Methods Chromatograpic separation was performed on Agilent InfinityLab Poroshell 120EC-C(4.6 mm×150 mm,3 μm) column with a gradient elution program with mixtures of methanol and 5 mmol ammonium acetate solution as the mobile phase at a flow rate of 0.4 mL·min^(-1).Q-TOF-MS was developed for throughput screening of the preliminary chemical profile of ginger Pinellia ternata and ginger Pinellia pedatisecta Schott in the positive ion mode.Profinder software found the compounds through MFE algorithm.Different compounds in Pinellia ternata and Pinellia pedatisecta Schott were determined,with the threshold P<0.01 and the minimum multiple change value FC<2.0,and the models were established.Results Totally 12 compounds specific to ginger Pinellia ternata and 99 components specific to ginger Pinellia pedatisecta Schott were found.The data models of ginger Pinellia ternata and ginger Pinellia pedatisecta Schott were established.The model could predict the sample deviation when some ginger Pinellia pedatisecta Schott palm was added.Conclusion The unique components of ginger Pinellia pedatisecta Schott can be determined by Q-TOF-M and MPP,the high content of which can be used to establish data model with certain predictability,providing ideas for the detection of adulteration in ginger Pinellia ternata.
