摘  要:为解析钙离子胁迫对槲蕨根茎生长影响的生理机制,以两年生槲蕨植株为实验材料,观察了钙离子胁迫对其根茎的影响。将温室培养的两年生槲蕨植株移栽至直径18.5 cm的花盆中,以蛭石为基质,置于光照培养箱,浇施霍格兰德营养液7 d后,分别浇施含有不同钙离子含量的霍格兰德营养液,处理14 d,根据槲蕨叶片的形态变化确定处理浓度。取0,600,1200 mmol·L^(-1)钙离子浓度处理的槲蕨根茎测定脯氨酸含量、甜菜碱含量、电导率含量、超氧阴离子含量、过氧化氢含量、抗坏血酸过氧化物酶活性、单脱氢抗坏血酸还原酶活性、脱氢抗坏血酸还原酶活性和谷胱甘肽还原酶活性。结果显示,根茎的甜菜碱的含量随着钙离子浓度的增加而升高,而脯氨酸的含量仅在1200 mmol·L^(-1)钙离子浓度时显著升高;电导率也是在1200 mmol·L^(-1)钙离子浓度时显著升高;过氧化氢含量先升高后降低,且在1200 mmol·L^(-1)钙离子浓度时仍高于对照组,而超氧阴离子的含量随着钙离子浓度的增加而升高;抗坏血酸过氧化物酶、单脱氢抗坏血酸还原酶和谷胱甘肽还原酶活性随着钙离子浓度的增加而增强,脱氢抗坏血酸还原酶活性仅在1200 mmol·L^(-1)钙离子浓度时显著升高。表明钙离子浓度在1200 mmol·L^(-1)以下时主要产生渗透胁迫作用。In order to analyze the physiological mechanism of calcium ion stress on rhizome growth of Drynaria fortune,the effects of calcium ion stress on rhizome growth of two years D.fortune were observed.The two-year-old plants were transplanted into a pot with a diameter of 18.5 cm in greenhouse.Vermiculite was used as the substrate and placed in a light incubator.Hoagland nutrient solution was applied for 7 days,and then Hoagland nutrient solution with different calcium ions content was applied for 14 days.Take 0,600,1200 mmol·L^(-1)calcium ion concentration processing D.fortune rhizome,and determine contents of proline,betaine,electrical conductivity,superoxide anion content,hydrogen peroxide content,ascorbic acid peroxidase activity,single dehydrogenation ascorbic acid reductase activity,dehydrogenation ascorbic acid reductase activity and glutathione reductase activity.The results showed that the content of betaine increased with the increase of calcium ion concentration,while the content of proline only increased significantly at 1200 mmol·L^(-1)calcium ion concentration.The electrical conductivity also increased significantly at 1200 mmol·L^(-1)calcium ion concentration.The content of hydrogen peroxide increased first and then decreased,and was still higher than that of the control group at 1200 mmol·L^(-1)calcium ion concentration,while the content of superoxide anion increased with the increase of the calcium ion concentration.The activities of ascorbate peroxidase,monodehydroascorbate reductase and glutathione reductase increased with the increase of calcium ion concentration,but the activity of dehydroascorbate reductase increased significantly only at 1200 mmol·L^(-1)calcium ion concentration.The results showed that calcium ion concentration below 1200 mmol·L^(-1)mainly produced osmotic stress.
