摘  要:为了不断改进左旋肉碱的合成工艺,研究了一种合成左旋肉碱中间体L-卡内腈的新路线。该路线以(R)-(-)-环氧氯丙烷为起始原料,先与液体氢氰酸发生加成反应得到(R)-(+)-4-氯-3-羟基丁腈;然后(R)-(+)-4-氯-3-羟基丁腈再与三甲铵盐酸盐和氨水反应得到产品L-卡内腈,副产品为氯化铵。该合成新路线的摩尔总收率可达到75%以上,合成的L-卡内腈化学纯度和手性纯度均较高,反应条件温和,过程绿色环保,适宜进行大规模的工业化生产。In order to optimized the synthetic process of L-carnitine,a new synthetic route of its intermediate L-carnitrile was studied.(R)-(-)-epichlorohydrin was used as the starting material,firstly,the addition reaction with liquid hydrocyanic acid to obtain(R)-(+)-4-chloro-3-hydroxybutanitrile;Then(R)-(+)-4-chloro-3-hydroxybutyronitrile is reacted with trimethylammonium hydrochloride and ammonia water to obtain the product L-carenonitrile,as a by-product of ammonium chloride.The total molar yield of L-carnesonitrile is more than 75%,which own high chemical purity and chiral purity,besides,the reaction conditions are mild and the process was green.Therefore,we can come to the conclusion that this new synthetic route of L-carnitrile is suitable for large-scale industrial production.
