摘 要:基于超高效液相色谱-串联质谱法(UPLC-MS/MS),建立了饲料中万古霉素和去甲万古霉素2种糖肽类抗生素的测定方法。样品经0.1%甲酸水-乙腈提取,MCX固相萃取柱净化,UPLC-MS/MS进行分析。方法学验证表明,万古霉素和去甲万古霉素在10~1000 ng/mL范围内线性关系较好(R>0.999),定量限分别为50.3,54.7μg/kg;在100,200,1000μg/kg加标水平下,平均回收率在79.3%~108.6%之间,相对标准偏差RSD在1.9%~9.8%(n=6)。该方法定量限低、精密度好,可应用于饲料中万古霉素和去甲万古霉素的定量分析。A method for determination of vancomycin and norvancomycin in animal feed was established by ultra-high pressure liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).The samples were extracted by 0.1%formic acid-acetonitrile and were purified on a MCX solid-phase extraction column.The samples were analyzed by UPLC-MS/MS.Methodological validation shows that good linearity(R>0.999)were observed in the range 10~1000 ng/mL for vancomycin and norvancomycin,the limit of quantitations were 50.3,54.7μg/kg.The recoveries ranged between 79.3%and 108.6%at three spiked(100,200,1000μg/kg)with relative standard deviations(n=6)between 1.9%and 9.8%.This method showed lower limit of quantification,good precision,and was suitable for the quantitative analysis of vancomycin and norvancomycin in animal feed.
