摘 要:目的:分析昼夜节律对肝脏脂代谢相关基因表达谱的影响,研究胆固醇代谢相关基因的昼夜节律性,以及高脂饮食、节律调节因子的缺失等因素对节律的影响。方法:选取小鼠肝脏组织昼夜节律表达谱的相关数据,利用CirGRDB数据库Lomb-Scargle周期表算法预测时间序列的周期,分析基因表达的昼夜节律性,并分析胆固醇代谢通路中具有昼夜节律性的基因,分析肝脏胆固醇代谢基因的昼夜节律变化。结果:小鼠肝脏中具有昼夜节律性的胆固醇代谢基因主要功能在于胆固醇合成、胆固醇排泄、胆汁酸的排泄、脂蛋白脂解酶的抑制,可能与动脉粥样硬化相关,高脂饮食对节律性影响较少,节律控制因子影响相关胆固醇代谢基因的节律。结论:肝脏中部分胆固醇代谢通路基因具有昼夜节律性,其节律性不受高脂饮食影响,节律紊乱可干扰肝脏胆固醇代谢的节律性。Objective:To analyze the effects of circadian rhythm on the expression profile of liver lipid metabolism-related genes and the circadian rhythm of cholesterol metabolism-related genes,as well as the effects of high-fat diet and the lack of rhythm regulators on the rhythm.Methods:The data related to the circadian rhythm expression profile of mouse liver tissue were selected.The circadian rhythm of gene expression,genes with circadian rhythm in cholesterol metabolism pathway and circadian rhythm changes of liver cholesterol metabolism gene were analyzed.All of these were calculated with the period of time series of Lomb-Scargle periodic table algorithm in CirGRDB database.Results:The circadian rhythm of cholesterol metabolism genes in mouse liver mainly functions in cholesterol synthesis,cholesterol excretion,bile acid excretion,and inhibition of lipoprotein lipolytic enzymes.High-fat diet has little effect on the rhythm of related genes,while the lack of rhythm control factors can lead to the loss of rhythm of related genes.Conclusion:Some of the hepatic cholesterol metabolism pathway genes have circadian rhythms,which affect the formation of atherosclerotic plaques,and rhythm disturbances interfere with the rhythm of liver cholesterol metabolism.
