摘 要:细菌感染已成为危害公共安全的最大威胁之一。光热疗法被认为是最有前途的抗菌策略,特别是对于多药耐药细菌的形成。通过酶模拟水合成法制备的水溶性聚(3,4-乙烯二氧噻吩):聚(苯乙烯磺酸钠)(poly(3,4-ethylenedioxythiophene):poly(p-styrenesulfonic acid),PEDOT:PSS),可以将808 nm的近红外能量转化为大量的热量,具有优异的光热效应,光热转化效率高达46.4%。与无机光热剂相比,水溶性PEDOT:PSS具有优异的生物相容性,为材料的非破坏性穿膜提供了前提。结合共聚焦荧光成像,发现PEDOT:PSS能够快速膜穿透进入细菌内部,与光热疗法相结合可破坏细菌内部重要生物活性分子(如蛋白质等),并导致细胞膜结构功能紊乱,进一步增强杀菌效率。可有效消除金黄色葡萄球菌(Staphylococcus aureus,S.aureus)和耐甲氧西林的金黄色葡萄球菌(Methicillin-resistant Staphylococcus aureus,MRSA)。Microbial contamination and super bacterial infection have become one of the biggest threats to public safety.Photothermal therapy has been considered as one of the most promising antibacterial strategies,especially for the formation of multidrug-resistant bacteria.In this paper,the water-soluble polymer[Poly(3,4-ethylenedioxythiophene):poly(p-styrenesulfonic acid),PEDOT:PSS]prepared by the enzyme-simulated water synthesis method can convert the near-infrared energy of 808 nm into a large amount of heat.It has the excellent photothermal effect,and the photothermal conversion efficiency is as high as 46.4%.Compared with inorganic photothermal agents,water-soluble PEDOT:PSS has excellent biocompatibility,which provides a prerequisite for non-destructive membrane penetration.It was found that PEDOT:PSS can quickly penetrate the membrane into the inside of the bacteria by the confocal fluorescence images.Combined with photothermal therapy,it can destroy the intracellular contents(such as proteins,etc.)of the bacteria,destroy the cell membrane structure,and further enhance the sterilization efficiency.It can effectively eliminate Staphylococcus aureus(S.aureus)and Methicillin-resistant Staphylococcus aureus(MRSA).It was prored that PEDOT:PSS has excellent antibacterial efficiency and potential application prospects.
