摘 要:探讨了酒石酸泰乐菌素与阳离子染料结晶紫在pH 2.4的Clark-Lubs缓冲溶液条件下相互作用的共振光散射图谱,建立了双波长叠加共振光散射技术(DWRLS)测定酒石酸泰乐菌素含量的新方法。实验研究发现,该体系在325 nm和491 nm处产生了两个强烈的特征散射峰。在波长325 nm处,酒石酸泰乐菌素在0.04~0.24 mg·L^(-1)内的质量浓度与共振光散射强度差值(ΔI_(RLS))呈线性关系,其线性回归方程为ΔI_(RLS)=19606C-787.4,相关系数R=0.9994,检出限为0.0059mg·L^(-1);在波长491 nm处,酒石酸泰乐菌素在0.04~0.24 mg·L^(-1)内的质量浓度与共振光散射强度差值(ΔI)呈线性关系,其线性回归方程为ΔI_(RLS)=13006C-449.07,相关系数R=0.9996,检出限为0.0055mg·L^(-1)。当采用双波长叠加共振光散射技术(DWRLS)测定时,酒石酸泰乐菌素在0.04~0.24 mg·L^(-1)内的质量浓度与共振光散射强度差值(ΔI_(RLS))呈线性关系,其线性回归方程为ΔI_(RLS)=32613C-1236.5,相关系数R=0.9998,其检出限为0.0028mg·L^(-1),是单波长检测法的2倍。所拟方法具备操作简便、灵敏度高等特点,成功地应用于酒石酸泰乐菌素含量的测定。The resonance light scattering spectrum characteristics of the interaction between tylosin tartrate and cationic dye crystal violet under Clark-Lubs buffer solution of pH 2.4 were discussed,and a new method of measuring the content of tylosin tartrate by two-wavelength resonance light scattering technology(DWRLS)was established.It is found that the system produces two strong characteristic scattering peaks at 325 nm and 491 nm.At the wavelength of 325 nm,the mass concentration of tylosin tartrate in the range of 0.04~0.24 mg·L^(-1)has a linear relationship with the difference of resonance light scattering intensity(ΔI_(RLS)).The linear regression equation isΔI_(RLS)=19606C-787.4,the correlation coefficient R=0.9994,and the detection limit is 0.0059 mg·L^(-1);At the wavelength of 491 nm,the mass concentration of tylosin tartrate in the range of 0.04~0.24 mg·L^(-1)has a linear relationship with the difference of resonance light scattering intensity(ΔI_(RLS)).The linear regression equation isΔI_(RLS)=13006C-449.07,the correlation coefficient R=0.9996 and the detection limit is 0.0055 mg·L^(-1).When the dual wavelength superposition resonance light scattering technique(DWRLS)is used for determination,the mass concentration of tylosin tartrate in the range of 0.04~0.24 mg·L^(-1)has a linear relationship with the resonance light scattering intensity difference(ΔI_(RLS)).The linear regression equation isΔI_(RLS)=32613C-1236.5,the correlation coefficient R=0.9998,and the detection limit is 0.0028 mg·L^(-1),which is twice that of the single wavelength detection method.The proposed method has the characteristics of simple operation and high sensitivity.It has been successfully applied to the determination of tylosin tartrate.
