摘  要:为了建立大肠杆菌脂多糖(lipopolysaccharide,LPS)体外诱导羔羊外周血单个核细胞炎症模型,该试验探究了不同作用时间(3 h、6 h、12 h)和不同作用浓度(0、0.016、0.08、0.2、0.4、1、2、4、6、8、10、50μg/mL)的大肠杆菌LPS溶液对羔羊外周血单个核细胞相对活力和炎症因子相对表达量的影响。结果表明:在处理时间小于12 h时,细胞相对活性没有显著差异,处理浓度高于6μg/mL时,细胞相对活性普遍低于对照组;在处理时间为6 h时,除0.4μg/mL以外其余各组炎症因子IL-1β的相对表达量高于3 h和12 h,均显著高于对照组(P<0.05);处理时间为3 h,处理浓度为6μg/mL时,IL-6的相对表达量显著高于对照组且达到峰值(P<0.01);当处理时间为6 h,处理浓度为4μg/mL时,IL-10的相对表达量极显著高于对照组且达到峰值(P<0.01);TNF-α的相对表达量在处理时间为6 h、处理浓度为1μg/mL时达到峰值且显著高于对照组(P<0.05)。综上所述,LPS处理时间为6 h时,各细胞炎症因子表达量普遍较高,结合细胞相对活力结果,应选取浓度为4μg/mL、处理时间为6 h建立LPS体外诱导的羔羊外周血单个核细胞炎症模型,用于后续试验。In order to establish an inflammatory model of lamb peripheral blood mononuclear cells induced by lipopolysaccharide(LPS)of Escherichia coli in vitro,this experiment explored the effects of E.coli LPS solutions with different treatment times(3,6,12 h)and different treatment concentrations(0,0.016,0.08,0.2,0.4,1,2,4,6,8,10,50μg/mL)on the relative viability and the relative expression of inflammatory factors of peripheral blood nuclear cells in lambs.The result showed that there was no significant difference in the relative viability of cells when the treatment time was less than 12 h,and the relative viability of cells was generally lower than that of the control group when the treatment concentration was higher than 6μg/mL.Except the treatment concentration of 0.4μg/mL,the relative expression of the inflammatory factor IL-1βat 6 h was higher than that at 3 h and 12 h,which was significantly higher than that in the control group(P<0.05).At a treatment time of 3 h and a treatment concentration of 6μg/mL,the relative expression of IL-6 was significantly higher than that of the control group and reached a peak(P<0.01).When the treatment time is 6 h and the treatment concentration was 4μg/mL,the relative expression of IL-10 was significantly higher than that of the control group and reached a peak(P<0.01).The relative expression of TNF-αwas highest when the treatment time was 6 h and the treatment concentration was 1μg/mL,which was significantly higher than the relative expression of the control group(P<0.05).In summary,when the LPS treatment time is 6 h,the expression of inflammatory factors in cells is generally higher.Combing with the relative viability of the cells,the treatment group whose treatment time was 6 h and treatment concentration was 4μg/mL should be selected to establish a lamb peripheral blood mononuclear cell inflammation model induced in vitro for subsequent study.
