摘  要:蛋白质的液-液相分离(Liquid-liquid separation of proteins,LLPS)是一种基本的生物物理现象,为生物结构的形成提供了机制,而LLPS的失调也会导致一系列的疾病,如白内障、镰状细胞性贫血和阿尔茨海默症。对于牛血清白蛋白(BSA),其溶液的相行为受到诸如具有拥挤效应的分子或盐等添加剂的影响。证明了三价金属盐YCl_(3)能够诱导BSA发生LLPS相行为。在YCl_(3)存在的溶液中,BSA会形成一个介于低临界盐浓度c^(*)和高临界盐浓度c^(*)之间的LLPS体系。由动态光散射实验得到的BSA电泳迁移率为LLPS短程吸引机制提供了直接证据,数据表明,LLPS过程中,Y^(3+)通过在BSA分子的负电荷残基之间形成阳离子桥而引起蛋白质分子之间的短程吸引导致BSA的LLPS相行为。Liquid-liquid separation of proteins(LLPS)is a fundamental biophysical phenomenon that provides a mechanism for the formation of biological structures,and disorders of LLPS can lead to a range of diseases,such as cataracts,sickle cell anemia,and Alzheimer's disease.In the case of bovine serum albumin(BSA),the phase behavior of its solution is affected by additives such as molecules with crowding effects or salts.In this study,it is proved that trivalent metal salt YCl_(3)can induce LLPS phase behavior in BSA.In the presence of YCl_(3),BSA will form an LLPS system between low critical salt concentration c and high critical salt concentration c^(*).The electrophoretic mobility of BSA from dynamic light scattering experiments provides direct evidence for the short-range attraction mechanism of LLPS.The data indicate that the short-range attraction between protein molecules is caused by the formation of cation Bridges between negatively charged residues of BSA molecules during LLPS,which leads to the LLPS phase behavior of BSA.
