摘 要:目的 探讨当归四逆汤通过调节氧化应激反应对衰老机体免疫功能的影响。方法 通过注射D-半乳糖(D-glactose, D-gal)制备亚急性衰老大鼠模型,实验设正常组、模型组、V组、当归四逆汤组,模型制备完成后给予当归四逆汤组大鼠灌胃相应剂量的当归四逆汤,V组大鼠给予维生素E,其余2组大鼠灌胃等容积0.9%氯化钠溶液。Morris水迷宫实验观察大鼠的学习记忆能力,测量大鼠脾、肝、胸腺的脏器指数,比色法检测大鼠血清中超氧化物歧化酶(superoxide dismutase, SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase, GSH-Px)、丙二醛(malondialdehyde, MDA),铁离子还原法检测大鼠血清中总抗氧化能力(total anti-oxidation capability, T-AOC),酶联免疫吸附试验检测大鼠血清中白细胞介素(interleukin, IL)-2、IL-6及IL-10水平。结果 与正常组比较,模型组大鼠进入逃生平台范围次数明显减少(P<0.01)、逃避潜伏期明显延长(P<0.01),脾脏、肝脏、胸腺指数皆明显降低(P<0.01),血清中SOD、GSH-Px、T-AOC、IL-2、IL-10含量明显降低(P<0.01),MDA、IL-6含量明显升高(P<0.01);与模型组比较,V组和当归四逆汤组进入逃生平台范围次数明显增多(P<0.01),逃避潜伏期明显缩短(P<0.05),脾脏、肝脏、胸腺指数皆明显升高(P<0.05,P<0.01),血清中SOD、GSH-Px、T-AOC、IL-2、IL-10含量明显升高(P<0.01)、MDA、IL-6含量明显降低(P<0.01)。与V组比较,当归四逆汤组大鼠血清中IL-2、IL-10含量明显升高(P<0.01),IL-6含量明显降低(P<0.01)。结论 当归四逆汤能够通过减弱衰老机体的氧化应激反应,提高免疫系统功能。Objective: To investigate the influence of Danggui Sini Decoction on the immune function of aging rats by regulating the oxidative stress response. Methods: Subacute aging rats model were prepared by injecting D-glactose(D-gal). Rats were randomly divided into normal group, model group, Vgroup and Danggui Sini Decoction group. After the modeling, rats in the Danggui Sini Decoction group were gavaged with the corresponding dose of Danggui Sini Decoction, rats in the Vgroup were given vitamin E, and rats in the other two groups were gavaged with equal volume of 0.9% NaCl.Morris water maze experiment was used to observe the learning and memory ability of rats.The organ index of the spleen, liver and thymus of rats was measured.Colorimetry was used to detect GSH-Px, MDA, SOD in rats serum.Ferric ion reducing antioxidant power was used to detect T-AOC in rats serum.Elisa was used to detect IL-2, IL-6 and IL-10 in rats serum.Results: Compared with the rats in the normal group, the number of times of rats in the model group entered the escape platform range was significantly reduced(P<0.01), and the escape latency was significantly prolonged(P<0.01),the indexes of spleen, liver and thymus were significantly reduced(P<0.01), the contents of SOD, GSH-Px, T-AOC, IL-2 and IL-10 in the serum were significantly reduced(P<0.01), the contents of MDA and IL-6 were significantly increased(P<0.01).Compared with the rats in the model group, the number of times of rats in the Vgroup and Danggui Sini Decoction group entered the escape platform range were significantly increased(P<0.01), and the escape latency was significantly shortened(P<0.05),the indexes of spleen, liver, and thymus were significantly increased(P<0.05,P<0.01), the contents of SOD, GSH-Px, T-AOC, IL-2 and IL-10 in the serum were significantly increased(P<0.01), the contents of MDA and IL-6 were significantly decreased(P<0.01).Compared with rats in the Vgroup, the serum contents of IL-2 and IL-10 in the Danggui Sini Decoction group were significantly increased(P<0.01
