摘 要:目的 优化黄芪生脉饮醇沉工艺,得出最优参数范围,用于指导生产操作。方法 采用P-B试验设计,以总多糖、黄芪甲苷、固含量为指标,考察醇沉前浸膏密度、乙醇浓度、含醇量、静置时间、搅拌速度、醇沉温度、搅拌时间对醇沉工艺的影响,得出醇沉工艺的关键影响因素为醇沉前密度、乙醇浓度和含醇量。采用Box-Behnken响应面法优化醇沉关键因素,用以指导生产操作。结果 确定最优醇沉工艺参数为醇沉前浓缩液密度1.26~1.28 g/ml,乙醇浓度85%,含醇量55%~63%,按最优工艺参数验证试验结果与预测值接近。结论 采用响应面法优化后的醇沉工艺用以指导生产操作可行。Objective To optimize the alcohol precipitation technology of Huangqi Shengmai Decoction and obtain the optimum technological parameters for guiding the production operation. Methods The P-B experimental design was used to investigate the influence of the density of the extract, ethanol concentration, alcohol content, standing time, stirring speed, ethanol precipitation temperature, and stirring time on the ethanol precipitation process with the total polysaccharide, astragaloside A, and solid content as indicators. It was concluded that the key factors affecting the ethanol precipitation process were the density before ethanol precipitation, ethanol concentration, and alcohol content. Box-Behnken response surface method was used to optimize the key factors of alcohol precipitation to guide production operation. Results The optimum preparation conditions of alcohol precipitation techniques of huangqi shengmai were as follows: the density of concentrated solution before alcohol precipitation was 1.26 ~ 1.28 g/ml, 85% ethanol was added to make the alcohol content reach 55% ~ 63%. The experimental results were close to the predicted values according to the optimum technological parameters. Conclusion The alcohol precipitation process optimized by response surface methodology is feasible to guide the production operation.
