摘 要:新近发现α1,3岩藻糖基转移酶 (FUT9)参与LeX 寡糖的合成。通过逆转录 聚合酶链式反应、免疫印迹分析和免疫组化等方法 ,研究了在不同激素状态下人子宫内膜FUT9基因的表达 ,以及LeX 寡糖表达的调控机制。结果显示 :FUT9基因在人子宫内膜中表达 ,且其mRNA在分泌期的表达水平高于增生期 ,口服米非司酮(mifepristone)后 ,增生期内膜的表达量增加 ;LeX 寡糖主要分布在人子宫内膜腔上皮和腺上皮细胞表面 ,在不同生理时期其变化趋势与FUT9基因相似 ,并且口服米非司酮后 ,分泌期内膜的表达量减少 ;在内膜中检测到 3种(33kD、35kD和 85kD)LeX 寡糖蛋白 ,口服米非司酮后 35kD的LeX 寡糖蛋白消失。结果表明 :FUT9基因在人子宫内膜中有明确表达 ,且其表达受卵巢激素的调节 ,孕激素对其有上调作用 ;卵巢激素在转录水平上实现对LeX 寡糖表达的调节。Recently it was found that α1,3-fucosyltransferase (FUT9) was an essential enzyme involved in the synthesis of LeX oligosaccharide. In this article, the expression ofFUT9 gene at different hormonal levels and the regulating mechanism of LeX synthesis in human endometrium were investigated by using RT-PCR, Immunohistochemistry staining and Western blotting.FUT9 mRNA was detected with human endometrium, and the levels of mRNA in secretory endometrium and in proliferative endometrium from patients who had taken mifepristone before operation, were higher than that of proliferative endometrium (P<0.01). LeX antigen was mainly located on the surface of epithelium. The floating level of LeX antigen expression was similar to that ofFUT9 gene. There were three (33 kD, 35 kD and 85 kD) LeX carrying proteins expressed in human endometrium, mifepristone treatment made 35 kD band disappear. The results indicate thatFUT9 gene is obviously expressed in human endometrium and can be upregulated by progesterone, and ovary hormones regulate the expression of LeX at the level of transcription.
