摘  要:目的观察熊果酸(UA)对大鼠海绵体神经损伤性勃起功能障碍(ED)的治疗作用及相关机制。方法选取18只8周龄雄性SD大鼠,随机分为假手术对照组(Sham)、模型对照组[双侧海绵体神经损伤(BCNI)]及治疗组(双侧海绵体神经损伤+熊果酸治疗),每组6只。大鼠双侧海绵体神经损伤后1 d,治疗组给予熊果酸灌胃治疗,假手术对照组和模型对照组均以等体积溶剂载体灌胃,4周后测定阴茎海绵体内压(ICP),应用马拉瑞改良法(Masson’s)三色染色检测阴茎海绵体组织平滑肌与胶原比值,免疫组织化学染色检测阴茎海绵体组织平滑肌含量,蛋白质印迹法(Western blot)检测阴茎海绵体组织α平滑肌肌动蛋白(α-SMA)和神经元型一氧化氮合酶(nNOS)表达水平。比较三组大鼠ICP最大值(ICPmax)、平均动脉压(MAP)、ICP最大值与MAP比值(ICPmax/MAP)、平滑肌/胶原比值、平滑肌含量及nNOS、α-SMA表达水平。结果假手术对照组ICPmax和ICPmax/MAP分别为(96±20)mm Hg(1 mm Hg=0.133 kPa)、(0.775±0.050),治疗组ICPmax和ICPmax/MAP分别为(70±12)mm Hg、(0.640±0.010),均明显高于模型对照组的(40±21)mm Hg、(0.392±0.010),差异具有统计学意义(P<0.05);三组MAP比较差异无统计学意义(P>0.05)。假手术对照组和治疗组平滑肌/胶原比值、平滑肌含量及nNOS、α-SMA表达水平均高于模型对照组,差异有统计学意义(P<0.05)。结论熊果酸能够通过抑制阴茎海绵体纤维化有效改善神经损伤性ED大鼠的阴茎勃起功能,而神经保护作用及抗氧化损伤作用可能是熊果酸对神经损伤性ED发挥治疗作用的潜在机制。Objective To observe the effect and mechanism of ursolic acid(UA)on erectile dysfunction(ED)induced by cavernous nerve injury in rats.Methods A total of 188-week-old male SD rats were selected and randomly divided into a sham-operated control group(Sham),a model control group[bilateral cavernous nerve injury(BCNI)]and a treatment group(bilateral cavernous nerve injury+ursolic acid treatment),with 6 rats in each group.After 1 d of bilateral cavernous nerve injury in rats,the treatment group was treated with ursolic acid gavage,and the sham-operated control group and the model control group were gavaged with an equal volume of solvent carrier.After 4 weeks,the intracavernosal pressure(ICP)of corpus cavernosum penis was measured,and the smooth muscle to collagen ratio of corpus cavernosum penis was measured by Masson’s trichrome staining,the smooth muscle content of penile cavernosal tissue was measured by immunohistochemical staining,and the expression levels ofα-smooth muscle actin(α-SMA)and neuronal nitric oxide synthase(nNOS)of corpus cavernosum penis were measured by protein blotting(Western blot).The ICP maximum value(ICPmax),ICPmax to mean arterial pressure(MAP)ratio(ICPmax/MAP),smooth muscle/collagen ratio,smooth muscle content and nNOS andα-SMA expression levels in the three groups of rats were compared.Results The ICPmax and ICPmax/MAP in the sham-operated control group were(96±20)mm Hg(1 mm Hg=0.133 kPa)and(0.775±0.050),and those in the treatment group were(70±12)mm Hg and(0.640±0.010),which were significantly higher than(40±21)mm Hg and(0.392±0.010)in the model control group,and the differences were all statistically significant(P<0.05).There was no statistically significant difference in MAP among the three groups(P>0.05).The smooth muscle/collagen ratio,smooth muscle content and nNOS andα-SMA expression levels in the sham-operated control group and treatment group were higher than those in the model control group,and the differences were statistically significant(P<0.05).Conclusion Ursolic
