摘 要:多糖是一类来源丰富、可降解和可再生的天然高分子,广泛应用于食品、医药、造纸、皮革等行业。然而,由于多糖结构中缺少可被特异性检测的基团,使得其在应用时的定性/定量分析较为困难。荧光标记技术能赋予多糖可被检测的荧光基团,是实现多糖应用可视化、进而研究多糖功能与作用机理的重要手段。文章综述了常见的荧光标记多糖的方法,包括在多糖的羟基、羧基和醛基引入荧光素的荧光标记法,以及通过化学反应构造共轭结构的自发荧光法,详细阐述了不同荧光标记法的原理、标记条件和应用效果。总结和展望了荧光多糖在荧光示踪、生物成像、食品监测、水质监测和智能包装材料等领域的应用。Polysaccharide is a kind of abundant,degradable and renewable natural macromolecules,which is widely used in food,medicine,papermaking,leather and other industries.However,the lack of specific detectable groups in molecular structures of polysaccharides results in the difficulty in their qualitative/quantitative analysis.Fluorescent labeling technology can endow polysaccharides with detectable fluorescent groups,which is an important way to achieve the visualization of polysaccharides during their application and further investigate their function and action mechanisms.This paper reviewed the common fluorescent labeling methods of polysaccharides,including the fluorescent labeling methods by introducing fluorescein into the hydroxyl,carboxyl and aldehyde groups of polysaccharides,and the autofluorescence method by constructing conjugated structures through chemical reactions.The principles,labeling conditions and applications of different fluorescent labeling methods were described in detail.The applications of fluorescent polysaccharides in the fields of fluorescent tracing,bioimaging,food monitoring,water quality monitoring and intelligent packaging materials were summarized and prospected.
