摘  要:目的通过气相色谱法检测经哈萨克族正常糖耐量人粪菌移植后的2型糖尿病db/db小鼠粪便中短链脂肪酸的含量,探讨肠道菌群代谢产物与糖尿病的关系。方法db/db小鼠分为模型组(db/db+PBS组)和哈萨克族正常糖耐量人粪菌液干预组(db/db+KNGT组),db/m小鼠为空白对照组(db/m+PBS组),每组9只,干预10周。分别在干预0周、6周和10周后检测小鼠空腹血糖、低密度脂蛋白胆固醇、高密度脂蛋白胆固醇、总胆固醇和三酰甘油的水平,并收集各组小鼠粪便,运用气相色谱技术检测干预0周、6周和10周后小鼠粪便中短链脂肪酸的含量。结果在哈萨克族正常糖耐量人粪菌液干预6周和10周后,与db/db+PBS组相比,db/db+KNGT组小鼠粪样中乙酸、丁酸的含量显著增加(F_(乙酸)=3.210,F_(丁酸)=8.530,均P<0.05)。结论哈萨克族正常糖耐量人粪菌液移植后小鼠粪样中菌群代谢产物乙酸和丁酸水平增加,从而改善了db/db小鼠的糖脂代谢紊乱。Objective To detect the content of short-chain fatty acids(SCFAs)in the feces of db/db mice after transplantation of fecal microbiota from Kazak donor with normal glucose tolerance(KNGT)by using gas chromatography(GC),and explore the relationship between intestinal flora metabolites and diabetes.Methods The db/db mice were divided into model group(db/db+PBS)and KNGT fecal microbiota transplantation intervention group(db/db+KNGT),9 mice in each group,and given orally 0.2 mL PBS or fecal microorganisms from a KNGT donor respectively for 10 weeks.At 0,6 and10 weeks after intervention,the levels of blood glucose,low-density lipoprotein cholesterol,high-density lipoprotein cholesterol,total cholesterol and triglyceride of mice were detected,and the feces of mice were collected to detect the content of SCFAs with GC.Results After 6 and 10 weeks of intervention,the contents of acetic acid and butyric acid in db/db+KNGT group significantly increased compared with db/db+PBS group(F_(acetic acid)=3.210,F_(butyric acid)=8.530,all P<0.05).Conclusion After the transplantation of KNGT fecal microbiota solution,the levels of acetic acid and butyric acid in the fecal samples of mice increased,thus improved the disorder of glucose and lipid metabolism in db/db mice.
