摘  要:通过系统性地研究脂多糖(LPS)对人原代角质细胞的细胞活性、IL-1α、IL-8及ROS等与皮肤炎症密切相关指标的影响,开发了一种基于人原代角质细胞的皮肤炎症模型。结果显示,0.01~100µg/mL LPS对人原代角质细胞活性没有较大影响,100µg/mL LPS能同时诱导人原代角质细胞产生大量IL-1α和IL-8,0.01µg/mL LPS能诱导产生大量ROS。因此,建立了对不同炎症指标使用LPS进行差异化诱导的人原代角质细胞皮肤炎症模型,同时用此模型对样品OSM2021041301进行检测,发现样品OSM2021041301能显著抑制LPS诱导升高的IL-1α、IL-8水平,而对LPS诱导升高的ROS水平有较弱的抑制作用,说明此样品对LPS及其类似物诱导的IL-1α、IL-8及ROS升高而引起的炎症具有一定的抗炎作用。Using inducers to induce cells to produce inflammatory response is a common in-vitro experimental method to study inflammation.However,relatively few inflammatory models have been developed for cosmetic industry,and there are great differences in model inducers,selection of inflammatory indicators,and inducer concentrations.Herein,a skin inflammation model based on human primary keratinocyte was developed,in which the effects of lipopolysaccharide(LPS)on the cell viability and the levels of IL-1α,IL-8 and ROS were systematically studied.The results showed that,0.01~100µg/mL LPS had no significant effect on the cell viability of human primary keratinocytes;100µg/mL LPS could simultaneously induce human primary keratinocytes to produce large amounts of IL-1αand IL-8;0.01µg/mL LPS could induce large amount of ROS.A skin inflammation model was thus established.Furthermore,sample OSM2021041301 was tested with this model.It was found that,sample OSM2021041301 could significantly inhibit LPS-induced elevated IL-1αand IL-8 levels,while its inhibitory effect on LPS-induced elevated ROS level was weak.It indicated that OSM2021041301 had certain anti-inflammatory effect on the inflammation caused by the increase of IL-1α,IL-8 and ROS induced by LPS and its analogues.
